    Abstract

    Stres merupakan respon fisiologi organisme yang disebabkan
    ketidaksesuaian lingkungan. Pada ikan gurame stres dapat diakibatkan oleh
    beberapa faktor salah satunya yaitu perbedaan suhu. Penelitian ini bertujuan
    untuk menyeleksi gen housekeeping pada ikan gurame (Osphronemus
    gouramy) yang diberi stres suhu rendah pasca pemberian suplemen
    Spirulina plantesis melalui identifikasi gen GAPDH (glyceraldehyde-3-
    phosphate dehydrogenase), 18S rRNA, dan ODC (Ornithin decarboxylase).
    Konsentrasi Spirulina plantesis yang digunakan yaitu 0%, 1%, dan 3% yang
    diberikan selama 40 hari untuk melihat suplemen dengan konsentrasi mana
    yang paling optimal dalam pertumbuhan ikan gurame. Pada akhir
    pemeliharaan ikan diberi stres berupa suhu rendah dibawah suhu yang
    seharusnya dia hidup yaitu pada suhu 20oC selama 5 menit untuk
    mengetahui resisteansi terhadap stres yang diberikan. Parameter yang
    diukur dalam penelitian ini yaitu tingkat kelulushidupan ikan, pertumbuhan
    ikan gurame meliputi panjang dan berat ikan. Kadar glukosa darah ikan
    diukur menggunakan alat glukosa kit one Touch. Darah ikan diuji pada jam
    ke 0, 2, 6, 12, dan 24 pasca stres suhu rendah. Kadar glukosa darah paling
    tinggi yaitu pada suplemen Spirulina plantesis konsentrasi 0% di jam ke 0
    sedangkan kadar glukosa yang stabil yaitu pada suplemen Spirulinan
    plantesis konsentrasi 3% kadar. Sampel dari setiap pengujian kadar glukosa
    akan diisolasi RNA untuk sintesis cDNA. Selanjutnya dilakukan analisa
    ekspresi gen housekeeping dengan cara amplifikasi pada cDNA organ hati.
    Hasil dari amplifikasi gen housekeeping menunjukkan adanya ekspresi padagen GAPDH (glyceraldehyde-3-phosphate dehydrogenase) dan 18S rRNA
    sedangkan pada gen ODC tidak terekspresikan sehingga tidak ada pita yang
    dihasilkan pada hasil elektroforesis gen tersebut. Hal ini menunjukkan
    bahwa gen housekeepeing GAPDH dan 18S rRNA dapat digunakan sebagai
    gen normalitas atau gen kontrol internal untuk menganalisa ekspresi gen;---Stress is a physiological response to an organism caused by an environmental mismatch. The stressed gouramy can be caused by several factors, one of which is temperature difference. This study aims to select housekeeping genes in gouramy (Osphronemus gouramy) which were given low-temperature stress after Spirulina plantesis supplementation through identification of GAPDH (glyceraldehyde-3-phosphate dehydrogenase), 18S rRNA, and ODC genes. Spirulina plantesis concentration used is 0%, 1%, and 3% given for 40 days to see which supplements are the most optimal in the growth and development of gouramy. At the end of the maintenance the fish was given stress in the form of a low temperature below that it was supposed to live at 20oC for 5 minutes to find out the resistance to stress given. Parameters seen in this study are fish survival and growth. Fish blood glucose levels were measured using a glucose one touch kit. Fish blood was tested at 0, 2, 6, 12, and 24 hours after low temperature stress. The highest blood glucose level was at 0% Spirulina plantesis supplement at 0 o'clock while the lowest glucose level was in Spirulina plantesis supplement concentration of 3%. Samples from each glucose level test will be isolated from RNA for cDNA synthesis. Next is housekeeping gene expression analysis by amplifying the liver organ cDNA. The results of the amplification of housekeeping genes showed the expression of GAPDH (glyceraldehyde-3-phosphate dehydrogenase) and 18S rRNA genes, while the ODC gene was not expressed so that there was no band produced on the electrophoresis of the gene. This shows that the housekeepeing GAPDH and 18S rRNA genes can be used as normality genes to analyze subsequent gene expression.
