Abstract

Penelitian yang berjudul “Sintesis dan Uji Katalepsi Nanopartikel Zn - Ekstrak Biji Karabenguk (Mucuna pruriens var.utilis) Indonesia” ini bertujuan untuk mengetahui karakteristik nanopartikel Zn – Mucuna pruriens (Zn-MPn) dan pengaruhnya dalam mengatasi katalepsi (rigiditas) pada mencit (Mus muculus) yang diinduksi haloperidol. Tahapan penelitian yang dilakukan terdiri atas ektraksi biji Mucuna pruriens; sintesis Zn-MPn; karakterisasi Zn-MPn dan uji katalepsi Zn-MPn. Ekstraksi biji Mucuna pruriens dilakukan melalui metode maserasi dengan menggunakan pelarut etanol:air (1:1) pH 3 (ditambahkan asam sitrat). Sintesis Zn-MPn menggunakan Zink asetat dihidrat (Zn(CH3COO)2.2H2O) dan ekstrak biji Mucuna pruriens (EMP). Karakterisasi Zn-MPn dilakukan dengan menggunakan instrumen FTIR, SEM, XRD dan TG-DTA. Uji katalepsi dilakukan pada Zn-MPn dosis 5, 15 dan 25 mg/kg berat badan. Hasil penelitian menunjukkan karakteristik Zn-MPn yaitu, adanya pita pada serapan 447,5 cm-1 dan 1342,4 cm-1 mengindikasikan ikatan Zn-O dan Zn-N serta pergeseran bilangan gelombang dan penurunan intensitas serapan gugus O-H, N-H, NH2, dan C-O dari EMP berdasarkan spektra FTIR. Zn-MPn berupa agregat seperti ditunjukkan oleh gambar SEM. Difraktogram x-ray menunjukkan Zn-MPn bersifat amorf dan keberadaan Zn pada Zn-MPn ditunjukkan pada posisi 2θ = 31,41º (sesuai JCPDS file 361451), serta diperkirakan memiliki ukuran partikel rata-rata 22,21 nm. Profil TG-DTA menunjukkan Zn-MPn memiliki kestabilan termal lebih baik dibandingkan EMP. Hasil uji katalepsi menunjukkan Zn-MPn mampu menurunkan katalepsi secara signifikan, dengan dosis 5 dan 15 mg/kg berat badan menurunkan katalepsi secara signifikan dibandingkan EMP 200 mg/kg berat badan. Diantara ketiga dosis, dosis yang paling baik menurunkan katalepsi adalah dosis 15 mg/kg berat badan. Kata Kunci : Mucuna pruriens, sintesis, nanopartikel, Zn-MPn, katalepsi The study entitled "Synthesis and Catalepsy Test of Zn - Indonesian Velvet Bean seeds (Mucuna pruriens var.utilis) Extract Nanoparticles" is aimed to dertermine the characteristics of Zn - Mucuna pruriens nanoparticles (Zn-MPn) and its influence to reduced catalepsy (in term of rigidity) in mice (Mus muculus) induced by haloperidol. The stages of this research involved extraction of Mucuna pruriens seeds; synthesis of Zn-MPn; characterization of Zn-MPn and catalepsy test of Zn-MPn. Extraction of Mucuna pruriens seeds was conducted using maceration method with ethanol:water (1:1) solvent pH 3 (citric acid added). Synthesis of Zn-MPn used Zinc acetate dihydrate (Zn(CH3COO)2.2H2O) and Mucuna pruriens seeds extract (EMP). Characterization of Zn-MPn was performed by means of FTIR, SEM, XRD and TG-DTA. Catalepsy test conducted for Zn-MPn doses level of 5, 15 and 25 mg/kg. The results showed the characteristics of Zn-MPn that is the presence of absorption band at 447,5 cm-1 dan 1342,4 cm-1 indicated for Zn-O and Zn-N bonds, wave numbers changes and decreasing absorption intensity of OH, NH, NH2, and CO groups from MPE based on FTIR spectra. The synthesized Zn-MPn formed aggregates as shown by SEM images. X-ray diffraction patterns showed that Zn-MPn was amorphous structure and the presence of Zn in the Zn-MPn in the position of 2θ = 31,41º (corresponding JCPDS file 361451), and Zn-MPn is estimated to have an average particle size of 22,21 nm. TG-DTA profile showed that Zn-MPn had a better thermal stability than EMP. The catalepsy test results showed Zn-MPn at three doses is able to reduce catalepsy significantly, with doses level of 5 and 15 mg/kg can reduce catalepsy significantly compared to EMP 200 mg/kg. Among the three dose level, the best dose to reduce catalepsy is a dose level of 15 mg/kg. Keywords: Mucuna pruriens, synthesis, nanoparticles, Zn-MPn, catalepsy.
