Abstract

Penelitian yang dilakukan bertujuan untuk mengevaluasi respons pucuk kentang (Solanum tuberosum L.) terhadap adanya cekaman salinitas dengan mengukur parameter morfologi dan fisiologi pucuk kentang secara in vitro pada medium MS 0 yang telah diberi penambahan konsentrasi NaCl (0, 50, 100 dan 150 mM). Pengukuran parameter morfologi yaitu multiplikasi pucuk, tinggi pucuk, dan jumlah nodus yang dilakukan setiap minggu selama empat minggu, sedangkan pengukuran parameter fisiologi yaitu kadar klorofil diukur dengan metode Arnon (1949) menggunakan spektrofotometer. Data diolah menggunakan rancangan acak lengkap dengan enam kali ulangan. Hasil penelitian menunjukkan bahwa cekaman salinitas memberi pengaruh terhadap semua parameter yang diukur. Kecepatan multiplikasi pucuk pada medium konsentrasi 50 mM NaCl lebih tinggi dibandingkan dengan kecepatan multiplikasi tanaman pada medium konsentrasi NaCl yang lainnya. Hasil untuk pertambahan nodus dan tinggi pucuk menunjukkan semakin tinggi konsentrasi NaCl, pertambahan nodusnya semakin sedikit dan ukuran tanaman pendek. Semakin tinggi konsentrasi NaCl juga menyebabkan kadar klorofil yang terkandung pada plantlet semakin sedikit. Penghambatan yang paling tinggi akibat adanya cekaman salinitas dari semua parameter yang diukur adalah pada konsentrasi 150 mM NaCl. The aim of this research was to evaluate the response of potato (Solanum tuberosum L.) shoot towards salinity stress with measure morphology and physiology of potato plant that was grown in vitro. The shoot were grown in MS 0 medium that was added by NaCl concentration (0, 50, 100 dan 150 mM). Morphology characteristics measured were shoot multiplication, shoot height and total nodes, in which it were measured every week for four weeks. Physiology characteristic measured was chlorophyll concentration which was carried out by Amon method (1949) using spectophotometry. Data was analyzed usingcompletely randomized design with six replications.Results suggested that salinity stress influenced all parameter. The rapidity of multipication in plant medium with 50 mM NaCl was higher than in other medium.Nodes accretionand chlorophyll concentration wereinfluenced by NaCl concentration; higher NaCl concentration resulted inlowernodes accretion and lower chlorophyll concentration. NaCl concentration that caused highest inhibition due to salinity stress was 150 mM NaCl concentration. Keyword: shoot multiplication, height, total nodes, chlorophyll concentration.
