Abstract
Penelitian ini bertujuan untuk mengekstraksi dan menganalisis senyawa dari berbagai bagian tumbuhan ciplukan (Physalisangulata).Tahapan penelitian yang dilakukan meliputi tahapan: (i) pemilihan kondisi ekstrak situmbuhan ciplukan yang meliputipemilihanjenispelarut, waktuekstraksidanjumlahekstraksi; (ii) ekstraksi bagian akar, batang, buah, dan daun ciplukan dengan menggunakan kondisiekstraksi yang telahdipilih; (iii) analisisprofilfitokimiamenggunakan HPLC dengandetektor UV; (iv) sertaidentifikasisenyawadalambagiantumbuhanciplukan yang mengandungsenyawa paling banyakdenganmenggunakan UPLC dengan detector massa ESI-QTOF. Hasil penelitian menunjukkan bahwa kondisi ekstraksi menggunakan ultrasonicator dengan pelarut etanol 100%, selama 30 menit, dandilakukansebanyaktiga kali memberikanhasilekstraksi paling tinggi. Analisisprofilfitokimiamenggunakan HPLC berbagaibagiantumbuhanciplukanmenunjukkanbahwabagiandauntumbuhanciplukanmengandungjumlahsenyawa yang lebihbanyakdenganintensitassenyawa polar yang lebih tinggi dibandingkan bagian tumbuhan lain. Analisis lebih lanjut terhadap bagian daun menggunakan UPLC-ESI-QTOF menunjukkan bahwa terdapat Sembilan puncak senyawa dalam ekstrak daun tumbuhan ciplukan.Berdasarkan data ion indukdanfragmentasinya, lima senyawaberhasildiidentifikasi, yaituduasenyawa flavonoid quercetin-3-O- rutinosidedan 3-O-metilquercetin-O-rutinoside, tiga senyawa with asteroiddarijenisphysalin, yaituphysalin D, physalinG, danphysalinF.;--- This research is to isolate and determine the phytochemicals of various parts of ciplukan plant (Physalisangulata). The reearch consist of four steps i.e.: (i) selection of extraction conditions of ciplukan plants, including the solvent; extraction duration, and a number of extraction; (ii) extraction of the roots, stems, fruits, and leaves parts of ciplukan plant using extraction conditions that have been determined; (iii) analysis of phytochemicals profile using HPLC with UV detector; (iv) identification of compounds in one of the extracts that contained more compounds using UPLC with mass detector ESI-QTOF. The results showed that the extraction conditions using ultrasonicator with 100% ethanol, for 30 minutes, and performed three times giving the highest extraction results. Analysis of HPLC-UV chromatogram showed that leaves contained more compounds than other part. The leaves extract also had higher intensity of polar compounds than others. Based on UPLC-ESI-QTOF, there were nine compounds in leaves extract, of which five of them were determined as quercetin-3-O-rutinoside, 3-O- metilquercetin-O-rutinoside, physalin D, physalin G, and physalin F.
