Abstract

Morinda citrifolia L. (mengkudu) mengandung sejumlah besar senyawa metabolit sekunder golongan fenolik dan antrakuinon. Daun pada M. citrifolia L. sering terbuang sia-sia dan belum banyak dimanfaatkan, namun diketahui mengandung senyawa fenolik dan antrakuinon yang memiliki aktivitas anti bakteri, anti-jamur, anti-virus, antitumor, analgesik, dan anti inflamasi. Penambahan zat pengatur tumbuh pada medium kultur in vitro tumbuhan digunakan untuk pembentukan dan pertumbuhan kalus yang menghasilkan senyawa fenolik dan antrakuinon. Zat pengatur tumbuh 2.4-D dan kinetin sering digunakan dalam induksi kalus karena peranannya dalam dediferensiasi sel dan pembelahan sel. Penelitian ini bertujuan untuk menganalisis kalus M. citrifolia L. hasil kultur eksplan daun yang ditanam pada medium MS (Murashige&Skoog) dengan menggunakan 12 kombinasi perlakuan zat pengatur tumbuh 2.4-D (1-1.75 mg/l) dan kinetin (1-1.5 mg/l). Hasil penelitian menunjukkan pertumbuhan kalus terbaik pada konsentrasi tertinggi yaitu kombinasi 2.4-D 1.75 mg/l dan kinetin 1.5 mg/l dengan hasil rerata pertambahan berat basah kalus sebesar 47.83 mg/hari. Kalus yang diinduksi pada medium perlakuan kombinasi 2.4-D dan kinetin lebih optimal memberikan respon pada proses pembelahan dan poliferasi sel serta dapat memberikan respon dalam proses morfogenesis sehingga mampu menghasilkan kalus kompak berwarna kuning kecokelatan yang mampu mengakumulasi senyawa metabolit sekunder. Hasil analisis kualitatif pada kalus dan daun segar M. citrifolia L. menggunakan uji fitokimia mampu mendeteksi adanya kandungan senyawa fenolik dan antrakuinon. Hasil penelitian ini menunjukkan bahwa dengan penambahan zat pengatur tumbuh 2.4-D dan kinetin efektif dalam merespon pertumbuhan kalus M. citrifolia L. dan dapat mengakumulasi senyawa fenolik dan antrakuinon.;--- Morinda citrifolia L. (noni) contains a large number of secondary metabolites from the group of phenolic and anthraquinone. The leaves on M. citrifolia L. is often wasted and has not been widely used, but it is known to contain phenolic and anthraquinone compounds which has anti-bacterial activity, anti-fungal, anti-viral, anti-tumor, analgesic, and anti-inflammatory. The addition of growth regulators on in vitro plant culture medium are used for callus formation and growth that produces phenolic and anthraquinone compounds. Growth regulator 2.4-D and kinetin are often used in callus formation due to its role in cell de-differentiation and proliferation. This research aimed to analyze the M.citrifolia L. callus cultures leaf explants grown on MS medium (Murashige and Skoog) using 12 combined treatment plant growth regulator 2.4-D (1-1.75 mg / l) and kinetin (1-1.5 mg/l). The results showed that the highest callus growth on concentrations of 2.4-D is a combination of 1.75 mg/l kinetin and 1.5 mg/l with an average gain of wet results callus of 47.83 mg/l day. Callus was induced on medium combination treatment of 2.4-D and kinetin more optimal response to the process of cell division and proliferation and gives response in morphogenesis process so as to produce a yellow-brown compact callus which were able to accumulate secondary metabolites. The results of qualitative analysis on the callus and fresh leaves of M. citrifolia L. using phytochemical test is able to detect the content of phenolic compounds and anthraquinone. The results showed that the addition of plant growth regulator 2.4-D and kinetin effectivelly to respond growth of callus M. citrifolia L. and were able to accumulate phenolic and anthraquinone compounds.
