Abstract
Mikroalga hijau-biru Spirulina sp. banyak dimanfaatkan pada bidang pangan dan kosmetik karena mengandung sejumlah pigmen dengan bioaktivitas yang beragam. Klorofil dan fikosianin merupakan dua pigmen dominan yang terkandung dalam Spirulina sp. Pada penelitian ini dilakukan karakterisasi dan studi fisikokimia fikosianin dan klorofil berupa pengaruh paparan cahaya, penambahan ion logam dan oksidan. Karakteristik klorofil dan fikosianin ditentukan menggunakan spektrofotometer UV-Vis dan FTIR, dan KLT untuk klorofil. Pengaruh paparan cahaya dilakukan dengan memberikan paparan lampu putih, kuning dan UV-C selama tujuh hari, kemudian diukur absorbansinya setiap 24 jam. Pengaruh penambahan ion logam Sn2+, K+, Fe3+, dan agen pengoksidasi H2O2 serta pereduksi Na2SO3 terhadap kestabilan klorofil dan fikosianin, dievaluasi dengan mengukur absorbansi masing-masing pigmen pada 0,5; 2; 5; dan 10 jam perlakuan. Analisis spektrofotometer menujukkan adanya puncak serapan pada panjang gelombang 662 nm dan 616 nm serta 620 nm yang menunjukkan keberadaan klorofil dan fikosianin. Hasil FTIR menunjukkan serapan khas gugus yang terdapat pada klorofil dan fikosianin. Hasil penelitian menunjukkan bahwa klorofil dan fikosianin tidak stabil terhadap cahaya ketiga lampu. Ion logam Sn2+, K+, dan Fe3+ serta agen pengoksidasi H2O2 dan pereduksi Na2SO3 dapat mengganggu kestabilan klorofil dan fikosianin, namun meningkatkan kestabilan fikosianin dengan tingkat kemurnian 1,43 terhadap paparan cahaya. ..... Green-blue microalgae Spirulina sp. widely used in the field of food and cosmetics because it contains a number of pigments with diverse bioactivity. Chlorophyll and phycocyanin are the two dominant pigments contained in Spirulina sp. In this study the characterization and physicochemical studies of chlorophyll and phicocyanin in the form of the effect of light exposure, addition of metal ions and oxidants. Characteristics of chlorophyll and phycocyanin were determined using UV-Vis and FTIR spectrophotometers, and TLC for chlorophyll. The effect of light exposure is done by giving exposure to white, yellow and UV-C lights for seven days then the absorbance is measured every 24 hours. The effect of metal ions Sn2+, K+, Fe3+ and H2O2 oxidizing agents and reducing Na2SO3 to the stability of chlorophyll and phycocyanin was evaluated by measuring the absorbance of each pigment at 0,5 ; 2; 5 and 10 hours of treatment. Spectrophotometer analysis showed that there were absorption peaks at wavelengths of 662 nm and 616 nm and 620 nm which indicated the presence of chlorophyll and phycocyanin. FTIR results show the typical absorption of clusters found in chlorophyll and phycocyanin. The results showed that chlorophyll and phycocyanin was not stable to all light of the lamp. Metal ions Sn2+, K+ and Fe3+ and H2O2 oxidizing agents and reducing Na2SO3 can interfere with the stability of chlorophyll and phycocyanin, but increase the stability of phycocyanin with purity level 1,43 to light exposure.
