Abstract

Penelitian tentang isolasi dan identifikasi bakteri pendegradasi senyawa fenol dari limbah cair industri kertas telah dilakukan. Penelitian ini bertujuan untuk mengisolasi dan mengetahui karakter bakteri yang mampu mendegradasi senyawa fenol dari limbah cair industri kertas. Penelitian ini dilakukan dalam 3 tahapan yaitu tahap isolasi bakteri, tahap identifikasi, dan tahap uji kemampuan isolat terpilih dalam mendegradasi senyawa fenol. Tahap isolasi bakteri dilakukan dengan menggunakan metode pengenceran, kemudian dilakukan pertumbuhan isolat dalam medium selektif yaitu Mineral Salt Medium (MSM) yang ditambahkan 0,26 mg/L larutan fenol. Tahap identifikasi dilakukan dengan berbagai tahapan pengujian yaitu karakterisasi morfologi, pewarnaan Gram, pewarnaan endospora, serta uji aktivitas biokimiawi. Selanjutnya identifikasi bakteri berpedoman pada buku Bergey’s Manual Of Systematic Bacteriology Ninth Edition (1994) dan Cowan and Steel’s Manualfor the Identification of Medical Bacteria Third Edition (1993). Hasil dari isolasi didapatkan 2 isolat yang berbeda, isolat pertama dinamakan dengan isolat limbah kertas 1 yang selanjutnya disingkat LK1 dan isolat kedua dinamakan dengan isolat limbah kertas 2 dan disingkat LK2. Kedua isolat yang ditemukan diduga memiliki kemiripan dengan genus Pseudomonas dan Staphylococcus. Hasil dari uji kemampuan isolat dalam mendegradasi fenol dengan metode analisis HPLC menunjukkan bahwa isolat LK1 dan isolat LK2 mampu mendegradasi fenol sebanyak 56,3 mg/L dalam rentang waktu 24 jam. Kata Kunci : Limbah Cair Kertas, Bakteri Pendegradasi Fenol, Senyawa Fenol, Karakterisasi Morfologi, Uji Biokimiawi, Analisis HPLC. Research on isolation and identification of bacteria degrading phenolic compounds from liquid waste paper industry has done. This study aimed to isolate and determine the character of the bacteria that can degrade phenolic compounds from liquid waste paper industry. The research was carried out in 3 stages: stage of bacterial isolation, identification phase and the test phase was selected isolates in degrading ability of phenolic compounds. Stage bacteria isolation performed using dilution method, and then conducted a selective growth medium that isolates the Mineral Salt Medium (MSM) added 0.26 mg / L of phenol solution. Phase identification is done by the various stages of testing the characterization of morphology, Gram staining, endospore staining, and biochemical activity assay. Further identification of bacteria based on the book Bergey's Manual of Systematic Bacteriology Of Ninth Edition (1994) and Cowan and Steel's Manual for the Identification of Medical Bacteria Third Edition (1993). The results obtained from the isolation of two different isolates, the first isolates is named isolate of the waste paper 1, and it called LK1 and the second isolate is isolate of waste paper 2 and it called LK2. Both of the founded isolates have allegedly similarities the Pseudomonas genus and the Staphylococcus genus. The results from isolate ability to degrade phenol test by HPLC analysis method showed that isolates LK1 and LK2 can degrade phenol as many as 56.3 mg / L in the span of 24 hours.
