Abstract

Penelitian mengenai potensi metabolit sekunder yang dihasilkan oleh beberapa isolat bakteri endofit akar tanaman obat Ageratum conyzoides dan Vetiveria zizanioides telah berhasil dilakukan. Tujuan dari penelitian ini ialah untuk mengetahui adanya aktivitas dari metabolit sekunder isolat bakteri endofit sebagai penghambat pertumbuhan jamur Trichophyton mentagrophytes. Lima isolat bakteri endofit yakni isolat O (Pseudomonas aeruginosa), I13 (Pantoea sp.), I14 (Klebsiella pneumonia), B14 (Staphylococcus equorum), dan B15 (Staphylococcus sp.) ditumbuhkan pada medium Luria Bertani (LB) cair hingga mencapai fase stasioner. Kultur bakteri disentrifugasi untuk memisahkan supernatan dan pelet. Supernatan digunakan dalam uji aktivitas anti jamur. Konsentrasi supernatan yang diuji ialah 20%, 40%, 60%, 80%, dan 100%. Konsentrasi didapatkan dengan cara mengencerkan supernatan menggunakan akuades steril. Uji aktivitas anti jamur dilakukan dengan metode broth microdilution menggunakan 96-well microtitration plate. Jumlah spora dari jamur uji kemudian dihitung menggunakan hemocytometer dan hasilnya dianalisis dengan uji Mann-Whitney U menggunakan SPSS 16.0 for windows. Hasil analisis data menunjukkan adanya perbedaan aktivitas penghambatan yang signifikan antara masing-masing kelompok perlakuan konsentrasi supernatan dari masing-masing isolat bakteri endofit. Supernatan isolat B14 (S. equorum) pada konsentrasi 100% memiliki aktivitas penghambatan terhadap T. mentagrophytes yang paling tinggi dan berbeda signifikan dengan 21 dari total 26 perlakuan termasuk kontrol.----------The research about potential of secondary metabolites from endophytic bacteria which isolated from Ageratum conyzoides and Vetiveria zizanioides as medicine plant has been conducted. The purpose of this research is to investigate antifungal activity of endophytic bacteria against Trichophyton mentagrophytes. Five endophytic bacteria isolates which are isolate O (Pseudomonas aeruginosa), I13 (Pantoea sp.), I14 (Klebsiella pneumonia), B14 (Staphylococcus equorum), and B15 (Staphylococcus sp.) were inoculated into Luria Bertani (LB) broth medium until stationer phase. Furthermore, each of bacteria culture were centrifugated to separate supernatant from pellet. Supernatant used in antifungal activity test. Supernatant concentration which used for antifungal activity test are 20%, 40%, 60%, 80%, dan 100%. These concentratrion made by diluting the supernatant with sterile aquadest. Antifungal activity test conducted with broth microdilution method using 96-well microtitration plate. The number of tested fungi spores then counted with hemocytometer and the result was analyzed with SPSS 16.0 for windows using Mann-Whitney U test. The result showed that each of supernatant concentration treatment from each endophytic bacteria isolates have different inhibition activity. Supernatant from isolate B14 (S. equorum) in 100% concentration has the highest inhibition activity to T. mentagrophytes and has different significance with 21 from total 26 treatments including control.
