Abstract

Physalis angulata atau dikenal sebagai “Ciplukan” di Indonesia termasuk kedalam familia Solanceae. Ciplukan yang berhabitus semak seringkali dimanfaatkan sebagai makanan dan obat tradisional. Variasi genetik dapat menyebabkan terjadinya perbedaan khasiat farmakologis, sehingga variasi genetik menjadi aaspek pertimbangan yang penting bagi masyarakat. Penelitian ini bertujuan untuk menganalisis variasi genetik populasi Ciplukan untuk mengetahui aksesibilitas masyarakat dalam memperoleh Ciplukan serta bertujuan untuk mengkonfirmasi data dari hasil penelitian sebelumnya. Popuplasi yang dianalisis berasal dari daerah Bandung yang diwakili oleh lima subpopulasi, yaitu Bandung Timur, Bandung Selatan, Bandung Barat, Bandung Utara, dan Bandung Tengah dengan total 23 individu sebagai sampel. Genom DNA diekstraksi dan diamplifikasi dengan metode RAPD (Random Amplified Polymorphisme DNA) menggunakan primer OPB18 dengan persentase polimorfisme 100% dan OPA16 dengan persentase polimorfisme 34,42%. Analisis klastering menggukan UPGMA (Unweighted Pair-Group Method with Aritmatic Average) dan PCA (Principal Component Analysis) menunjukan tidak terdapat pengelompokan populasi ciplukan berdasarkan lokasi asal Ciplukan. Hasil tersebut diperkuat dengan perolehan nilai aliran gen antar populasi Ciplukan di Bandung cukup tinggi (Nm = 1,6863). Aliran gen yang tinggi dapat mencegah terjadinya diferensisi akibat hanyutan gen. Analisis variasi genetik ciplukan perlu dilakukan dalam skala yang lebih luas guna menjadi dasar strategi pemanfaatan ciplukan yang lebih luas. ---------- Physalis angulata otherwise known as "Ciplukan" in Indonesia belongs to the family of Solanceae. Ciplukan is a shurb often used as food and traditional medicine. Genetic variation can lead to differences in pharmacological properties, so that genetic variation becomes an important consideration for society. This study aims to analyze the genetic variation of Ciplukan population to know the accessibility of the community in obtaining Ciplukan and aims to confirm data from previous research results. The analyzed popuplasi came from the Bandung area represented by five subpopulations, namely East Bandung, South Bandung, West Bandung, North Bandung, and Central Bandung with a total of 23 individuals as a sample. The DNA genome was extracted and amplified by RAPD (Random Amplified Polymorphism DNA) method using primer OPB18 with percentage of 100% polymorphism and OPA16 with percentage of polymorphism 34,42%. The clustering analysis using UPGMA (Unweighted Pair-Group Method with Aritmatic Average) and PCA (Principal Component Analysis) showed no grouping of population Ciplukan based on the origin location of Ciplukan. These results are reinforced by the acquisition of gene flow values among Ciplukan population in Bandung is quite high (Nm = 1,6863). High gene flow can reduce the fluid diferensiasi due to gene drift. Analysis of Ciplukan genetic variation needs to be done on a wider scale in order to form the basis for a wider Ciplukan utilization strategy.
