Abstract

Penelitian ini bertujuan untuk mengeksplorasi metode yang efektif dan efisien untuk mengekstraksi lipid dari mikroalga Botryococcus braunii menggunakan pelarut organik dan cairan ionik cis-oleil imidazolinium asetat. Tahapan penelitian dimulai dengan sub kultur Botryococcus braunii, persiapan biomassa, ekstraksi lipid menggunakan pelarut organik diantaranya heksan, heksan:metanol (7:3); metanol dan metanol:kloroform (1:2), ekstraksi menggunakan cairan ionik menggunakan 2 jenis ko-pelarut yaitu metanol dan metanol:kloroform (1:2) dan analisa profil lipid menggunakan instrumen GC-MS. Persentase ekstrak lipid pada penggunaan pelarut organik adalah 52,05% menggunakan metanol sedangkan penggunaan heksan:metanol (7:3), heksan, dan metanol:kloroform (1:2) masing –masing menunjukan 31,04%, 18,40% dan 17,59%. Pada penggunaan cairan ionik persentase ekstrak lipid adalah 73,83% menggunakan ko-pelarut metanol dan 18,40% menggunakan ko-pelarut metanol:kloroform (1:2). Profil lipid yang berbeda teramati pada kedua metode tersebut. Profil lipid pada fraksi metanol menunjukan profil yang paling beragam dibandingkan dengan penggunaan pelarut organik lainnya, dengan mengesktrak enam jenis asam lemak yaitu asam palmitat, asam palmitooleat, asam stearat, asam linoleat, asam linolenat, dan asam eikosanoat. Pada penggunaan pelarut cairan ionik dan ko-pelarut metanol menunjukkan profil lipid yang paling beragam dibandingkan penggunaan pelarut lainnya. Berdasarkan hasil penelitian yang telah dilakukan, penggunaan cairan ionik pada proses ekstraksi pelarut dapat meningkatkan perolehan ekstrak lipid dari mikroalga Botryococcus braunii.;---This study aims to explore the effective and efficient method for extracting lipids from microalgae Botryococcus braunii using organic solvent and ionic liquid cis-oleil imidazolinium acetate. The study involved in the sub culture of Botryococcus braunii, preparation of biomass, and lipid extraction using an organic solvent such as hexane, hexane: methanol (7:3); methanol and methanol: chloroform (1:2), and extraction using ionic liquids with methanol and methanol: chloroform (1:2) as ionic liquid cosolvent. The effective and efficient extraction was evaluated by comparing the weight ratio of extracted lipid to the biomass applied, while the lipid profiles was clarified by GC-MS analysis. The percentage lipid extract analysis of organic solvent show that methanol extracted 52,05%, while hexane:methanol (7:3), hexane and methanol:chloroform (1:2) are 31.04%; 18,04%; and 17,59% respectively. In addition, an ionic liquid with methanol extracted 73.83% and 18,40% using methanol:chloroform (1:2) as ionic liquid cosolvent. The different profile of lipid was observed between both of the method. Lipid profile in methanol fraction of organic solvent extraction method is more diverse than methanol combined with hexane and chloroform. It was monitored six type of fatty acid which are palmitic acid, palmitooleic acid, stearate acid, linoleic acid, linolenic acid, and eicosanoic acid. In addition, the more variation of lipid extract content was observed using ionic liquid with methanol as cosolvent. Based on this research, it can be conclude that applying ionic liquid in the solvent extraction process may increase the extract lipids mass from microalgae.
