Abstract
Beberapa pohon P. duclitan di Kebun Raya Bogor tumbuh dengan kondisi dan tempat yang sama namun mengalami perbedaan pada bentuk buah, sehingga diperlukan klarifikasi untuk menentukan status taksonomi tanaman tersebut. Tujuan penelitian ini adalah mengetahui hubungan kekerabatan secara fenetik antara beberapa tanaman sawo P.duclitan dan membandingkannya dengan P. obovata dan P. campechiana sebagai spesies pembanding berdasarkan data RAPD (Random Amplified Polymorphic DNA). Primer OPB-17 mengamplifikasi DNA Pouteria dengan baik. Tingkat polimorfisme yang dihasilkan oleh primer OPB-17 bernilai 65,38% yang berarti tingkat polimorfismenya tinggi, sehingga dapat digunakan dalam analisis fenetik. Analisis fenetik menggunakan software MEGA 4 berhasil mengklasifikasikan beberapa tanaman sawo Pouteria yang ditandai dengan adanya dua kelompok yang terbentuk. Hasil ini menunjukkan adanya perbedaan dan indikasi varietas baru pada P. duclitan terutama P. duclitan yang didukung oleh data morfologi yaitu buah yang lonjong dan bulat. Selain itu ada P. duclitan yang membentuk kelompok sendiri yang diduga berasal dari tetua yang jauh hubungan kekerabatannya dengan tetua varietas lain. Kata kunci : Pouteria, Fenetik dan RAPD Some of Pouteria duclitan trees at Bogor Botanical Garden can grow in the same condition and place but it has distinction in fruit form, thus it needs clarification to determine their status. The aim of this research is to know the phenetic relationship among samples of P. duclitan, and compare it with P. obovata and P. campechiana as comparision species, based on RAPD (Random Amplified Polymorphic DNA) data. Primer OPB-17 amplyfied species of Pouteria with good result. The degree of polymophisme of this primer was 65,38%, meaning that its degree was high, thus it could be used in phenetic analysis. The phenetic analysis used Mega 4 software, successfully classify some plants of Pouteria in to two groups. This result showed that there were some defferences, indicating that there were new variety of P. duclitan. It’s supported by the data of morphology with fruit in spindle and spherical form. Beside that, one of P. duclitan made different group, and is hypothesized coming from different ancestors. Keyword : Pouteria, Phenetic and RAPD
