Abstract

Indonesia memiliki berbagai jenis kacang-kacangan lokal yang memiliki kandungan nutrisi yang baik dan berpotensi menjadi pangan fungsional namun masih belum banyak dieksplorasi kandungannya termasuk diantaranya adalah kacang tunggak, kacang gude, kacang lurik dan kacang beras. Namun selain bernilai gizi baik, di dalam kacang-kacang ini ditemukan adanya senyawa antinutrisi yang dapat menurunkan bioavaibilitas dan kecernaan nutrisi dalam tubuh. Penelitian ini berfokus pada analisis kandungan metabolit pada kacang-kacangan tersebut dengan menggunakan pendekatan metabolomik untargeted menggunakan instrumentasi UHPLC-ESI-QTOF. Data metabolomik diproses menggunakan open source software MS-DIAL, KnowItAll, dan dianalisis statistik menggunakan MetaboAnalyst 5.0. Analisis kemometrik dilakukan dengan metode SAM, PCA, PLSDA, dan HCA. Berdasarkan hasil analisis multivariat VIP diketahui terdapat 15 senyawa metabolit dari 12 golongan senyawa yang berbeda dapat membedakan sampel kacang. Berdasarkan model statistik PLS-DA dan analisis VIP, sampel kacang beras dapat dibedakan oleh kandungan senyawa golongan benzenoida, sampel kacang lurik dibedakan oleh senyawa golongan asam organik dan turunan, sampel kacang gude dibedakan dari senyawa golongan non-logam homogen, dan sampel kacang tunggak dibedakan oleh golongan senyawa fenilpropanoid dan poliketida. Diketahui bahwa terdapat dua senyawa antinutrisi yang teridentifikasi dalam sampel yaitu Punikafolin dan Tellimagrandin II. Senyawa ini termasuk dalam kelompok tannin terhidrolisis yang terkandung dalam sampel kacang. Selain itu juga diketahui hubungan kekerabatan antara empat jenis sampel kacang, yaitu sampel kacang tunggak memiliki hubungan kekerabatan paling dekat dengan sampel kacang beras, dan kacang gude memiliki hubungan kekerabatan dengan kacang lurik. Hubungan kekerabatan ini diperoleh dari hasil klastering berupa dendrogram dan profil metabolit yang ditampilkan melalui heatmap dan hasilnya menunjukkan kesesuaian dengan filogeni ke empat kacang tersebut. Indonesia has various types of lockal beans that have good nutritional content and have the potential to become functional foods but their content has not been explored much, including cowpeas, pigeon pea, lurik peas and black eyed pea. However, apart from having good nutritional value, these peas also contain anti-nutritional compounds that can reduce the bioavailability and digestibility of nutrients in the body. This study focuses on the analysis of the metabolite content in these nuts using an untargeted metabolomics approach using the UHPLC-ESI-QTOF instrumentation. Metabomic data were processed using the open source software MS-DIAL, KnowItAll, and statistically analyzed using MetaboAnalyst 5.0. Chemometric analysis was performed using SAM, PCA, PLSDA, and HCA methods. Based on the results of VIP multivariate analysis, it was found that there were 15 metabolites that could distinguish the peanut samples analyzed, including groups of coumarin glycosides, trialkylamine, phosphatidicin, fatty acid esters, butenolides, methylpidine, thienopyridine, biphenyl and their derivatives, styrene, phenylpropane, dialkyl ether, non-metallic halides, tricarboxylic acids and their derivatives prenyl sulfoxide, and monoterpenoids. Based on the VIP analysis, black-eyed peas samples can be distinguished by the content of compounds in the benzenoide group, lurik peas samples are distinguished by organic acids and derivatives, pigeon peas samples are distinguished from homogeneous non-metallic compounds, and cowpea samples are distinguished by phenylpropanoid and polyketide compounds. It is known that there are two antinutritional compounds identified in the sample, namely Punicafolin and Tellimagrandin II. This compound belongs to the group of hydrolyzed tannins contained in the peanut sample. In addition, it is also known that there is a kinship relationship between four types of peanut samples, namely cowpea samples have the closest relationship with black-eyed peas samples, and pigeon peas have a kinship relationship with lurik peas. This kinship was obtained from the results of clustering in the form of dendrograms and metabolite profiles displayed through a heatmap and the results showed conformity to the phylogeny of the four beans.
