Abstract
Penelitian ini bertujuan untuk mengetahui pengaruh kopigmentasi antosianin esktrak kulit manggis dengan campuran ion logam Fe(III) dan alginat terhadap efek hiperkromik, pergeseran batokromik, kandungan total antosianin monomerik, aktivitas antioksidan dan stabilitas termal. Penelitian ini dilakukan dengan cara ekstraksi kulit buah manggis menggunakan etanol 70% selama 24 jam. Ekstrak yang sudah dikeringkan dengan cara freeze drying dikopigmentasi dengan campuran ion logam Fe(III) dan alginat dengan variasi ekstrak antosianin kulit manggis 1g/L, FeCl3 0,0025M, dan alginat 0,1% masing-masing pada perbandingan (40:0:0), (40:3:0), (40:4:0), (40:5:0), (40:0:5), (40:3:5), (40:4:5), dan (40:5:5) dalam buffer asetat pH 4. Hasil kopigmentasi dilakukan pengukuran panjang gelombang maksimal, kandungan total antosianin monomerik dengan metode perbedaan pH, aktivitas antioksidan dengan metode DPPH dan stabilitas pemanasan pada suhu 600C ±20C. Dihasilkan rendemen antosianin ekstrak kulit manggis sebanyak 7,33% dan mengandung 9,1677 mg/L antosianin. Pada uji panjang gelombang maksimal semakin banyak jumlah ion logam Fe(III) yang ditambahkan semakin tinggi efek hiperkromik dan pergeseran batokromik yang dihasilkan. Penambahan alginat dapat meningkatkan efek hiperkromik namun menurunkan pergeseran batokromik hasil kopigmentasi antosianin dengan ion logam Fe(III). Penambahan ion logam Fe(III) dan alginat menurunkan kandungan total antosianin monomerik dan aktivitas antioksidan. Hasil kopigmentasi dengan penambahan alginat pada kopigmentasi antosianin dengan logam Fe(III) dapat mempertahankan stabilitas termal pada suhu 600C ±20C dibandingkan dengan yang ditambahankan ion logam Fe(III) saja atau yang tanpa kopigmentasi; This study aims to determine the effects copigmentation of anthocyanins in mangosteen peel extract with a mixture of Fe(III) metal ions and alginate on the effects of hyperchromic, batochromic shift, total anthocyanins content, antioxidant activity and thermal stability. This research was carried out by extracting mangosteen peel using 70% ethanol for 24 hours. The dried extract by freeze drying was copigmented with mixture of Fe(III) metal ions and alginate with variations of mangosteen peel anthocyanins extract 1g / L, FeCl3 0,0025M, and alginate 0.1% in each ratio (40:0:0), (40:3:0), (40:4:0), (40:5:0), (40:0:5), (40:3:5), (40:4:5) , and (40:5:5) in acetate buffer pH 4. The results of the copigmentation was measured by maximum wavelength, the total anthocyanins content was measured by pH difference method, antioxidant activity was measured by DPPH method and thermal stability was measured at 60°C ± 2°C. The anthocyanins content of mangosteen peel extract was 7,33% and contained 9,1677 mg/L anthocyanins. In the maximum wavelength test, the more amount of Fe(III) metal ions were added, the higher the hyperchromic effect and the batochromic shift will be. The addition of alginate to the copigmentation of anthocyanins with Fe(III) metal ions can increase the hyperchromic effect but reduce the batochromic shift. The more amount of Fe(III) metal ions and alginate were added, the anthocyanins content and antioxidant activity will decrease. The results of copigmentation with the addition of alginate to the copigmentation of anthocyanins with Fe(III) metal ions can maintain the thermal stability at 60°C ± 2°C compared to the addition of Fe(III) metal ions only or those without copigmentation.
