Abstract

Bakteri patogen pada ikan merupakan suatu masalah dalam bidang perikanan. Pemberian antibiotik sintetik secara terus menerus mengakibatkan timbulnya resistensi baru terhadap bakteri. Senyawa metabolit sekunder pada tumbuhan dapat dimanfaatkan sebagai antibakteri. Penelitian mengenai pengaruh ekstrak kulit akar tumbuhan mangrove Avicennia alba dan Rhizophora apiculata telah berhasil dilakukan. Tujuan penelitian ini untuk mengetahui pengaruh ekstrak dan konsentrasi optimal dari ekstrak Avicennia alba dan Rhizophora apiculata terhadap pertumbuhan bakteri Pseudomonas aeruginosa, Salmonella typhi, dan Staphylococcus aureus. Ekstraksi menggunakan pelarut etanol pro analyts. Ekstrak kulit akar A. alba dilarutkan dalam DMSO 100% (Dimetyl sulfoxide) dan R. apiculata dilarutkan dalam DMSO 10%. Kedua ekstrak dibuatkan konsentrasi yang sama yaitu 2%, 4%, 8%, 16%, dan 20%. Pengujian aktivitas antibakteri menggunakan metode disc-diffusion. Desain penelitian menggunakan Rancangan General Faktorial dengan 5 kali pengulangan. Tetracyclin 0,01% sebagai kontrol positif, DMSO 10% dan DMSO 100% sebagai kontrol negatif. Hasil pengujian ekstrak kulit akar A. alba pada konsentrasi 20% memiliki zona hambat tertinggi terhadap bakteri S. aureus (15,9 mm ± 4,204) mm, ekstrak kulit akar R. apiculata dengan konsentrasi 20% memiliki zona hambat tertinggi terhadap bakteri S. aureus (12,3 mm ± 0,273) mm. Berdasarkan analisis statistika, pemberian ekstrak A. alba dan R. apiculata berpengaruh terhadap zona hambat yang dihasilkan terhadap bakteri. ;---Pathogenic bacteria in fish are a problem in the field of fisheries. The continuous provision of synthetic antibiotics results in new resistance to bacteria. Secondary metabolite compounds in plants can be used as antibacterials. Research on the effect of Avicennia alba and Rhizophora apiculata root bark extract has been successfully carried out. The purpose of this study was to determine the effect of extract and optimal concentration of extract Avicennia alba and Rhizophora apiculata on the growth of Pseudomonas aeruginosa, Salmonella typhi, and Staphylococcus aureus bacteria. Extraction using ethanol pro analyts. A. alba root bark extract was dissolved in 100% DMSO (Dimetyl sulfoxide) and R. apiculata dissolved in 10% DMSO. Both extracts were made with the same concentration of 2%, 4%, 8%, 16%, and 20%. Antibacterial activity testing uses the disc-diffusion method. The research design uses a General Factorial Design with 5 repetitions. Tetracyclin 0.01% as a positive control, DMSO 10% and DMSO 100% as a negative control. The test results of A. alba root bark extract at a concentration of 20% had the highest inhibition zone against S. aureus bacteria (15.9 mm ± 4.204) mm, R. apiculata root bark extract with a concentration of 20% had the highest inhibition zone against S. aureus bacteria (12.3 mm ± 0.273) mm. Based on statistical analysis, the administration of A. alba and R. apiculata extract affected the inhibitory zone produced against bacteria.
