Abstract
Penggunaan gula fermentasi yang semakin meningkat menyebabkan kebutuhan gula dan bahan baku ikut meningkat pula. Enzim sebagai biokatalisator dalam mendegradasi biopolimer di bidang industri dan pertanian telah banyak dilakukan untuk efesiensi biaya produksi gula. Selulase menjadi salahsatu enzim yang cukup diandalkan digunakan untuk mendegradasi polimer karbohidrat. Bahan berselulosa seperti jerami sangat melimpah di alam terbukti dapat didegradasi oleh Mikroorganisme melalui enzim yang dihasilkannya. Penelitian ini bertujuan untuk mengisolasi bakteri pendegradasi selulosa dari usus rayap Cryptotermes sp. dan mengidentifikasi jenisnya serta mengetahui karakterisasi enzim yang dihasilkan. Ada 4 tahapan utama dalam penelitian ini yaitu, isolasi bakteri dari usus rayap Cryptotermes sp., seleksi bakteri selulolitik, identifikasi secara biokimia dan aktivitas selulolitik yang dihasilkan. Hasil dari isolasi didapatkan 12 isolat kultur campuran, kemudian dilakukan uji screening pada medium CMC agar, 6 isolat mampu menghasilkan enzim selulase. Berdasarkan uji identifikasi menunjukkan kesamaan sifat biokimia diantaranya 2 isolat genus Clostridium, 2 isolat lagi Xenorhabdus dan 2 isolat terakhir yaitu Proteus dan Streptococcus. Aktivitas enzim selulase pada serbuk jerami yang terukur masing-masing spesies tersebut secara berurutan yaitu 0,883 U/ml, 0,558 U/ml), 0,591 U/ml, 0,785 U/ml, 0,121 U/ml dan 0,669 U/ml. pH optimum aktivitas selulase yaitu 6 pada empat isolat, sedangkan sisanya yaitu pH 5 dan 7. Salahsatu genus Clostridium memiliki kemampuan degradasi tertinggi dalam mengurai selulosa jerami padi;--- The increasing use of fermented sugar causes the need for sugar and raw materials to increase as well. Enzymes are biological catalysts in degrading biopolymers by industry and agriculture. Cellulase is one of the enzymes used to degrade carbohydrate polymers. Carbon fiber material is proven to be degraded by microorganisms through its enzymes. This becomes an interesting potential knowing rice straw is very abundant in nature. The aim of this research was to isolate the cellulose degrading bacteria from Cryptotermes sp. to identify its type and know the characterization of the enzyme produced. The steps of this study consisted of isolation of bacteria from termites gut Cryptotermes sp., Selection of cellulolytic bacteria, biochemical identification and cellulolytic activity produced. Among 12 isolates of bacteria, six isolates were cellulolytics. Based on the identification test had shown the similarity of the biochemical characteristic of the six isolates with the genus including Two isolates were identified in the genus of Clostridium, Two isolates were identified in the genus of Xenorhabdus, and the last two isolates in the genus of Proteus and Streptococcus. The measured activity of cellulase enzyme in media containing rice straw powder of each species was respectively 0.883 U / ml, 0,558 U / ml), 0,591 U / ml, 0,785 U / ml, 0,121 U / ml and 0,669 U / ml. These results indicate that Clostridium has the highest degradation ability in breaking rice straw cellulose
