Abstract

Studi mengenai potensi anti jamur dari isolat bakteri endofit akar tanaman Vetiveria zizanioides dan Ageratum conyzoides telah dilakukan. Tujuan dari penelitian ini adalah untuk menganalisis isolat bakteri endofit dan konsentrasi yang optimum untuk menghambat pertumbuhan khamir C. albicans. Satu isolat merupakan bakteri endofit akar tanaman V. zizanioides (isolat O) dan empat isolat lainnya merupakan bakteri endofit akar tanaman A. conyzoides (isolat B14, B15, I13 dan I14). Lima isolat bakteri endofit yang sudah disubkultur dari cryo preservation, dibiakan pada medium LB Broth hingga mencapai fase stationernya. Kultur kelima bakteri endofit kemudian disentrifugasi dan kemudian supernatannya diambil. Supernatan kelima isolat bakteri endofit kemudian diujikan pada C. albicans dengan menggunakan metode Broth microdilution, dengan konsentrasi 20, 40, 60, 80, 100 % (v/v). Pengaruh masing-masing supernatan dari isolat bakteri endofit diketahui dengan metode turbiditas, yang kemudian dikonversi ke dalam jumlah koloni dengan melalui persamaan kurva baku. Kelima isolat diketahui memiliki perbedaan yang signifikan terhadap kelompok perlakuan kontrol. Tiga isolat (isolat B15, O dan isolat I14) diantaranya menunjukkan penghambatan yang lebih besar dibandingkan dua isolat lain. Hal tersebut dikarenakan perbedaan karakteristik dari masing-masing senyawa metabolit sekunder dari masing-masing isolat.---------- Research about potential antifungal compounds of endophytic bacteria of Vetiveria zizanioides and Ageratum conyzoides root has been done. The purpose of research was analyze isolates of endophytic bacteria and optimum concentrations to inhibit the growth of Candida albicans. One isolate is endophytic bacteria from V. zizanioides (isolate O) and four isolates is endophytic bacteria from A. conyzoides (isolates B14, B15, I13 and I14). Five isolates endophytic bacteria have been subcultured from cryo preservation, cultured on LB Broth medium till reach the stationary phase. Then culture of the five endophytic bacteria was centrifuged and then the supernatant was taken. Then the supernatant of fifth isolates endophytic bacterial tested on C. albicans by using the Broth microdilution method, with concentrations 20, 40, 60, 80, 100% (v / v). The effect of each supernatant from isolates endophytic bacteria is known by turbidity method and converted into the number of colonies using the standard curve equation. The five isolates have significant differences to the control group. Three isolates (isolates B15, O and isolate I14) showed a greater inhibition than the two isolates. This differences causes by different mechanism and characteristic of antifungal compounds of each isolates.
