Abstract

Penelitian mengenai potensi metabolit sekunder yang dihasilkan oleh beberapa isolat bakteri endofit akar tanaman obat Ageratum conyzoides dan Vetiveria zizanioides telah berhasil dilakukan. Tujuan dari penelitian ini ialah untuk mengetahui adanya aktivitas dari metabolit sekunder isolat bakteri endofit sebagai penghambat pertumbuhan jamur C.albicans dan T.mentagrophytes. Sebanyak tujuh isolat bakteri endofit dan dua spesies jamur patogen dipelajari fase pertumbuhannya. Tujuh isolat bakteri endofit ditumbuhkan pada medium Luria Bertani (LB) cair hingga mencapai fase stasioner. Kultur bakteri disentrifugasi untuk memisahkan supernatan dan pelet. Supernatan digunakan dalam uji aktivitas antifungi. Konsentrasi kultur bakteri ditentukan dengan cara menimbang masing-masing supernatan dan pelet. Konsentrasi kultur bakteri yang diuji sebagai antifungi ialah 10 mg/ml dan 20 mg/ml. Uji aktivitas antifungi dilakukan dengan metode difusi cakram, dengan nystatin dan ketokonazol sebagai kontrol positif serta Akuades steril sebagai kontrol negatif. Zona hambat yang terbentuk diukur menggunakan jangka sorong dan hasilnya dianalisis dengan uji LSD dan Mann-Whitney menggunakan SPSS 16.0 for windows. Hasil analisis data menunjukkan adanya perbedaan aktivitas penghambatan yang signifikan antara masing-masing supernatan dengan konsentrasi kultur yang berbeda. Supernatan dari isolat B15 (Pseudomonas sp.) dan H (Pantoea sp.) dengan konsentrasi 10 mg/ml memiliki aktivitas penghambatan yang paling besar berturut-turut terhadap C.albicans dan T.mentagrophytes, sedangkan isolat B14 (Shewanella sp.) dan O (Pseudomonas aeruginosa) dengan konsentrasi 20 mg/ml memiliki aktivitas penghambatan yang paling besar berturut-turut terhadap C.albicans dan T.mentagrophytes.;---The research about potential of secondary metabolites from endophytic bacteria which is isolating from Ageratum conyzoides and Vetiveria zizanioides as medicine plant has been conducted. The purpose of this research is to investigate antifungal activity of endophytic bacteria to C.albicans and T.mentagrophytes. Growth curve of seven endophytic bacteria and fungal pathogens was identified. Seven isolates endophytic bacteria were inoculated into Luria Bertani (LB) broth medium until stationer phase. Furthermore, each of bacteria culture were centrifugated to separated supernatant from pellet. Supernatant used in antifungal activity test. Weight of supernatant and pellet of bacteria culture were measured with analytical balance in order to determine concentration of bacteria culture. All of concentration set as 10 mg/ml and 20 mg/ml. Test of antifungal activity used nystatin and ketokonazol as positive control and sterilized Aquadest as negative control with disk diffusion method. Inhibition zone measured with calipers and the result was analyzed with SPSS 16.0 for windows used LSD and Mann-Whitney test. The result showed that each of supernatant with different culture concentration have different inhibition activity. Supernatant from isolate B15 (Pseudomonas sp.) and H (Pantoea sp.) are the best to inhibit C.albicans and T.mentagrophytes in 10 mg/ml, while isolate B14 (Shewanella sp.) and O (Pseudomonas aeruginosa) are the best to inhibit C.albicans and T.mentagrophytes in 20 mg/ml.
