Abstract

Pola hidup tidak sehat, kurang berolahraga dan konsumsi asupan makanan yang tidak seimbang menjadi faktor hiperlipidemia. Hiperlipidemia adalah keadaan dimana darah terlalu tinggi mengandung kadar kolesterol total, Low Density Lipoprotein (LDL), dan trigliserida. Tujuan penelitian ini adalah untuk mengetahui adanya pengaruh pektin buah jeruk medan (Citrus sinensis cv. Medan) terhadap perbaikan lipid darah mencit jantan hiperlipidemia. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dan lima kali pengulangan dengan sampel penelitian adalah 30 ekor mencit jantan yang dibagi menjadi enam kelompok (kontrol negatif, kontrol positif, kadar pektin 1,5 g, kadar pektin 3 g, kadar pektin 4,5 g atau kadar pektin 6 g). Setelah dilakukan induksi pakan berlemak hingga mengalami hiperlipidemia mencit diberi perlakuan dengan cara menambahkan pektin dengan kadar yang berbeda-beda ke dalam 100 gram pakan berlemak selama 30 hari. Sampel darah diambil dari bagian vena caudalis dan dilakukan pengujian kadar lipid darah menggunakan metode CHOD-PAP, GPO-PAP, dan Formula Friedwald. Data yang didapatkan dianalisis dengan uji One Way Anova (p<0.05) dengan taraf kepercayaan 95%. Hasil penelitian ini menunjukkan bahwa pektin buah jeruk medan berpengaruh secara signifikan terhadap penurunan kadar trigliserida serum darah mencit dan peningkatan HDL serum darah mencit. Pektin buah jeruk tidak berpengaruh secara signifikan terhadap penurunan kolesterol dan LDL, namun pemberian pektin tetap dapat menurunkan rata-rata kadar kolesterol dan LDL setelah 30 hari pemberian pektin buah jeruk. Kadar pektin yang paling optimal dalam perbaikan lipid darah mencit adalah 6 gram / 100 gram pakan berlemak. Unhealthy lifestyle, lack of exercise, and unbalanced food consumption become hyperlipidemia factors. Hyperlipidemia is a condition where the blood contains high concentration of total cholesterol content, Low Density Lipoprotein (LDL), and triglycerides. The aim of the research is to find out the effect of medan citrus (Citrus sinensis cv. Medan) pectin on the blood lipid improvement of hyperlipidemia male mice. The research design was Completely Randomized Design (CRD) and five repetitions with the samples are 30 male mice divided into six groups (negative control, positive control, 1.5 grams pectin, 3 grams pectin, 4.5 grams pectin, and 6 grams pectin). After induction of fatty feed up until hyperlipidemia detected, the mice were treated by adding of pectin into 100 grams of fatty feed for 30 days. The blood sample was taken from vena caudalis part and blood lipid content was tested by CHOD-PAP method, GPO-PAP method, and Friedwald formula. Collected data were analyzed by One Way Anova test (p<0.05) with reliability of 95%. The results showed that citrus pectin given significantly effects on triglyceride content reduction and HDL improvement on blood serum of the mice. On the other hand, citrus pectin treatment showed no significantly effects on the improvement of cholesterol and LDL. However, citrus pectin still can reduce the average of content of cholesterol and LDL after 30 days treatment. The most optimal pectin content in the improvement of blood lipid of mice is pectin content of 6 grams / 100 grams fatty feed.
