Abstract
Artocarpus elasticus merupakan salah satu tumbuhan dari famili Moraceae yang telah diketahui kaya akan kandungan senyawa metabolit sekunder seperto golongan flavonoid. Flavonoid yang ditemukan dari famili Moraceae memiliki ciri khas yaitu flavonoid terprenilasi. Senyawa dari golongan flavonoid dilaporkan memiliki aktivitas biologis seperti antioksidan, antimikroba dan antidiabetes, selain itu juga flavonoid memiliki aktivitas sebagai inhibitor enzim asetilkolinesterase pada gangguan neurodegradatif seperti penyakit alzeimer, tetapi belum adanya penelitian senyawa flavonoid dari Artocarpus elasticus. Olehkarena itu, penelitian ini bertujuan untuk mengisolasi dan mengkarakterisasi senyawa flavonoid terprenilasi dari kayu batang Artocarpus elasticus serta potensinya sebagai inhibitor enzim asetikolinesterase. Metode isolasi dilakukan secara bertahap diantaranya ekstraksi dengan maserasi, pemisahan dan pemurnian dengan kromatografi cair vakum (KCV). Penentuan struktur senyawa murni dilakukan menggunakan analisis UV-Vis, FTIR, 1D NMR, 2D NMR dan MS. Pengujian aktivitas secara in silico dan in vitro. Hasil didapatkan dari tahapan isolasi dan karakterisasi diperoleh campuran resemat siklomorusin dari isolat 1 (114,1 mg), sikloartokarpin dari isolat 2 (50,5 mg), artokarpin dari isolat 3 (825 mg) dan kudraflavon C dari isolat 4 (78 mg). Hasil pengujian secara in silico menunjukkan bahwa isolat berinteraksi dengan enzim asetilkolinesterase dengan nilai energi afinitas ikatan pada (R)-siklomorusin sebesar -2,5 kkal/mol dengan 3 interaksi ikatan, (S)-siklomorusin sebesar -4,8 kkal/mol dengan 7 interaksi ikatan, sikloartokarpin sebesar -2,8 kkal/mol dengan 5 interaksi ikatan, artokarpin sebesar -4,0 kkal/mol dengan 8 interaksi ikatan, dan kudraflavon C sebesar -3,9 kkal/mol dengan 8 interaksi ikatan. Hasil pengujian secara in vitro didapatkan nilai %inhibisi campuran resemat siklomorusin sebesar 27,52%, sikloartokarpin tidak adanya inhibisi, artokarpin sebesar 48,32%, dan kudraflavon C sebesar 37,19% dibandingkan dengan kontrol galantamin sebesar 92,74%. Berdasarkan hasil penelitian ini, dapat disimpulkan bahwa senyawa flavonoid terprenilasi dari kayu batang Artocarpus elasticus berpotensi sebagai inhibitor enzim asetilkolinesterase meskipun termasuk inhibitor non kompetitif. Artocarpus elasticus is a plant from the Moraceae family that is known to be rich in secondary metabolites such as flavonoids. Flavonoids found in the Moraceae family are characterized by prenylated flavonoids. Compounds from the flavonoid group are reported to have biological activities such as antioxidant, antimicrobial, and antidiabetic properties. Additionally, flavonoids exhibit activity as acetylcholinesterase enzyme inhibitors in neurodegenerative disorders like Alzheimer's disease; however, no studies have been conducted on flavonoids from Artocarpus elasticus. Therefore, this study aims to isolate and characterize prenylated flavonoids from the wood of Artocarpus elasticus and assess their potential as acetylcholinesterase enzyme inhibitors. The isolation method was carried out in stages, including extraction by maceration, separation, and purification using vacuum liquid chromatography (VLC). The structure of the pure compounds was determined using UV-Vis, FTIR, 1D NMR, 2D NMR, and MS analyses. The activity was tested in silico and in vitro. The results obtained from the isolation and characterization stages yielded a mixture of cyclomorusin from isolate 1 (114.1 mg), cycloartocarpin from isolate 2 (50.5 mg), artocarpin from isolate 3 (825 mg), and kudraflavon C from isolate 4 (78 mg). In silico testing results showed that the isolates interacted with the acetylcholinesterase enzyme, with binding affinity energy values of -2.5 kcal/mol for (R)-cyclomorosin with 3 bond interactions, (S)-cyclomorosin at -4.8 kcal/mol with 7 bond interactions, cycloartokarpin at -2.8 kcal/mol with 5 bond interactions, artokarpin at -4.0 kcal/mol with 8 bond interactions, and kudraflavon C at -3.9 kcal/mol with 8 bond interactions. In vitro testing results showed that the inhibition percentage of the cyclomorusin racemic mixture was 27.52%, cycloartocarpin showed no inhibition, artocarpin was 48.32%, and kudraflavon C was 37.19% compared to the galantamine control at 92.74%. Based on these results, it can be concluded that the terpenylated flavonoids from the bark of Artocarpus elasticus have potential as acetylcholinesterase inhibitors, although they belong to the non-competitive inhibitor.
