    Abstract

    Resistensi antibiotik menjadi tantangan terbesar di Indonesia. Karena itu, diperlukan eksplorasi terhadap alternatif alami yang berpotensi sebagai senyawa antibakteri. Indonesia dikenal sebagai negara dengan kekayaan dan keanekaragaman hayati yang tinggi, termasuk pemanfaatan tanaman sebagai obat tradisional. Bunga Telang (Clitoria ternatea L.) merupakan salah satu tanaman obat dengan potensi antibakteri. Pemanfaatan senyawa metabolit sekunder bakteri endofit yang bersimbiosis dalam C. ternatea dapat menjadi alteratif untuk mencegah penurunan biodiversitas tanaman di Indonesia. Tujuan penelitian ini adalah mengisolasi dan mengetahui aktivitas antibakteri ekstrak supernatan bakteri endofit akar C. ternatea terhadap bakteri Staphylococcus aureus dan Escherichia coli. Penelitian diawali dengan isolasi bakteri endofit dari akar C. ternatea, diikuti dengan seleksi isolat potensial, identifikasi mikroskopis dan makroskopis, pengukuran kurva tumbuh, ekstraksi, serta pengujian aktivitas antibakteri dengan metode Disk Diffusion Assay (DDA), uji Minimum Inhibitory Concentration (MIC), uji Minimum Bactericidal Concentration (MBC), dan uji Time-Kill. Penelitian ini membuktikan bahwa ekstrak supernatan bakteri endofit dari akar C. ternatea memiliki aktivitas antibakteri terhadap S. aureus dan E. coli. Penelitian ini diharapkan mampu menambah wawasan mengenai aktivitas antibakteri ekstrak supernatan bakteri endofit akar C. ternatea sebagai sumber senyawa antibakteri alami, serta menjadi peluang untuk penelitian lebih lanjut mengenai karakterisasi dan mekanisme aktivitas senyawa antibakteri. Antibiotic resistance is the biggest challenge in Indonesia. Therefore, is necessary to explore alternative natural materials that have potential as antibacterial compounds. Indonesia known as a country that has high biodiversity, including in the utilization of plants as traditional medicine. Butterfly pea (Clitoria ternatea L.) is one of the medicinal plants that has potential as an antibacterial. Secondary metabolite compounds of endophytic bacteria symbiotic to C. ternatea can be an alternative to prevent the decrease of plant biodiversity in Indonesia. The purpose of this study was to determine the antibacterial activity of the supernatant extract of C. ternatea root endophytic bacteria against Staphylococcus aureus and Escherichia coli. The research began with the isolation of endophytic bacteria from the roots of C. ternatea, followed by selection of potential isolates, microscopic and macroscopic identification, growth curve measurement, extraction, and antibacterial activity test with Disk Diffusion Assay (DDA), Minimum Inhibitory Concentration (MIC) Assay, Minimum Bactericidal Concentration (MBC) Assay, and Time-Kill Assay. This study proves that the supernatant extract of endophytic bacteria from C. ternatea roots has antibacterial activity against S. aureus and E. coli. This research is expected to add insight into the antibacterial activity of C. ternatea root endophytic bacterial supernatant extract as a source of natural antibacterial compounds, as well as opportunities for further research on the characterization and mechanism of antibacterial compound activity.
