Abstract

The breadfruit plant (Artocarpus altilis) is one type of plant that shows potential as a source of medicinal plants. Breadfruit cultivation is widespread in various regions, where geographical differences and the ecosystem of each region can influence the content of secondary metabolites in a plant. Several groups of compounds that have been identified from the non-polar fraction of breadfruit plant extracts show the presence of steroids, terpenoids, flavonoids, carbohydrates, and fatty acids. Apart from that, a search of the literature shows that breadfruit plants have interesting biological activities, including anticancer, anti-inflammatory, antitubercular, and antiplasmodial. This research aims to determine the potential of breadfruit stem wood extract from West Java as an antioxidant and the content of secondary metabolites contained in the nonpolar fraction of the extract. The method used to test antioxidant activity is the DPPH method. Next, secondary metabolite compounds are isolated and characterized through several stages, namely extraction, fractionation, and purification. This fractionation and purification stage is carried out using various chromatography techniques, including Vacuum Liquid Chromatography, Gravity Column Chromatography, Radial Chromatography, and Thin Layer Chromatography. To identify the structure of compounds, FTIR and NMR spectroscopy are used. The research results show that the methanol extract of breadfruit plants has quite active potential as an antioxidant with an IC50 value of 79.36 μg/mL. Furthermore, the results of separation and purification of the nonpolar fraction of this extract produced one isolate with a mass of 59.9 mg in the form of a yellow sticky solid. FTIR data shows that isolate 1 has an absorption for the O-H group at 3444.98 cm-1, C-H sp3 at 2939.61 cm-1, C=O at 1720.56 cm-1, C=C aromatic at 1456.3 cm-1, C-H at 1371.4 cm-1, C-O at 1253.77 cm-1 and 1012.6 cm-1 which is typical for the flavonoid group. However, based on the results of NMR spectroscopy, it was discovered that the isolate obtained was still contaminated with fatty acids.
