Abstract
Studi mengenai deteksi gen anti fungi (surfaktin) pada isolat bakteri endofit dari akar tumbuhan obat Ageratum conyzoides L. dan Vetiveria zizanioides L. telah dilakukan. Tujuan dari penelitian ini adalah untuk mendeteksi dan menganalisis gen surfaktin dari tujuh isolat bakteri endofit dari akar tumbuhan tersebut. Tiga isolat (kode M, O dan H) merupakan bakteri endofit dari V. zizanioides dan empat isolat (kode I13, I14, B14 dan B15) merupakan bakteri endofit dari A. conyzoides. Penelitian ini dilakukan untuk mendapatkan informasi dan pemahaman mengenai gen anti fungi dan dapat digunakan untuk kepentingan penemuan dan produksi senyawa anti fungi. Tujuh isolat DNA bakteri endofit yang telah diisolasi DNA kromosom, diamplifikasi menggunakan metode PCR dengan primer yang memiliki target domain surfaktin sintase tioesterase sub unit dengan amplikon 441 bp. Segmen DNA yang telah teramplifikasi disikuensing dan dianalisis dengan metode bioinformatika. Setiap amplikon dianalisis untuk homologi dengan perangkat lunak BLASTX. Pohon filogenetik sikuen gen surfaktin dihasilkan dengan MEGA 7.0. Dalam penelitian ini, domain konservatif dideteksi dari sikuen oleh perangkat lunak Conserved Domain Search (CDS). Lima isolat menunjukan memiliki segmen gen surfaktin. Analisis homologi menunjukan segmen gen surfaktin yang didapatkan termasuk ke dalam superfamilia Abhidrolase. Berdasarkan analisis filogenetik gen surfaktin bakteri endofit akar homolog terhadap gen surfaktin dari kelompok Firmicutes. Selain itu berdasarkan analisis filogenetik mengindikasikan gen surfaktin isolat bakteri endofit A. conyzoides cenderung mengelompok sedangkan isolat bakteri endofit V. zizanioides terpisah dari kelompok bakteri endofit A. conyzoides. Hal tersebut menunjukan adanya keragaman gen surfaktin pada bakteri endofit akar A. conyzoides dan akar V. zizanioides.;--- Studies of detection antifungal gene (surfactin) in several isolates of endophytic bacteria from the roots of medicinal plants Ageratum conyzoides L. and Vetiveria zizanioides L. has been done. The purpose of this research was detected and analysed surfactin gene from seven bacterial endophytes isolates of plants root. Three isolates (code M,O and H) is bacterial endophytes from V. zizanioides and four isolates (code I13, I14, B14 and B15) is bacterial endophytes from A. conyzoides. This research was studied and analysed the antifungal gene bacteria endophytes. Moreover, it could discovered and produced an antifungal compounds. Seven samples of bacteria’s DNA chromosomes have been isolated and amplified using PCR with specific primers which targeting surfactin synthase thioesterase subunit domain with 441 bp DNA. Segment was sequenced and analyzed using bioinformatic methods. Each amplicons were analyzed for its homology with BLASTX software. Phylogenetic tree of surfactin gene sequences were generated with MEGA 7.0. Conservative domain was detected from sequence by Conserved Domain Search (CDS) software. The result of this research has showed that five isolates has surfactin gene segment. Analysis of homology showed that surfactin gene segment was included into superfamilia Abhidrolase. Based on phylogenetic analysis approach, surfactin gene from roots endophytes are homologous to the genes of the Firmicutes group. Phylogenetic analysis indicated that surfactin gene from A. conyzoides endophytes was grouping while surfactin gene from V. zizanioides endophytes was separated from group A. conyzoides endophytes. This research shows the diversity of surfactin gene in A. conyzoides and V. zizanioides.
