Abstract
Bacillus merupakan kontaminan umum makanan, seperti sayuran dan buah-buahan. Penelitian sebelumnya menyebutkan bahwa B. subtilis dapat menyebabkan pembusukan makanan dan B. cereus bertindak sebagai patogen makanan. Kedua bekteri ini merupakan bakteri pembentuk spora, yang memungkinkan mereka untuk bertahan hidup untuk waktu yang lama di bawah kondisi lingkungan yang merugikan. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antibakteri ekstrak biji kapulaga terhadap sel vegetatif dan spora B.cereus dan B. subtilis. Dalam penelitian ini, aktivitas antibakteri MIC dan MBC ekstrak metanol biji kapulaga diuji terhadap B. cereus ATCC33019 dan B.subtilis strain ATCC6633 menggunakan metode standar Clinical and Laboratory Standard Institute (CLSI). Hasil menunjukkan bahwa ekstrak biji kapulaga dapat menghambat sel-sel vegetatif B. cereus dan B.subtilis dengan Minimum Inhibitory Concentration (MIC) keduanya 1,25 mg/ mL. Sel-sel vegetatif B. cereus dapat dibunuh dengan Minimum Bactericidal Concentration (MBC) sebesar 1,25 mg/mL dan B. subtilis dengan MBC 10 mg/mL. Aktivitas Antispora diuji terhadap spora B. cereus dan B. subtilis kapulaga pada konsentrasi 0%, 1% dan 2% dengan waktu inkubasi 0 jam dan 1 jam. Ekstrak mereduksi jumlah spora B. cereus dari 5.16 menjadi 2.37 (CFU/mL) dan secara signifikan dipengaruhi oleh konsentrasi. Sementara itu, terhadap spora B.subtilis adalah 4,99 menjadi 2.32 dan signifikan dipengaruhi oleh waktu inkubasi. Secara keseluruhan, ekstrak methanol biji kapulaga menunjukkan potensi antimikroba dan antispora terhadap sel vegetatif dan spora B. cereus dan B. subtilis ----------- Bacillus is common contaminants of food, such as vegetables and fruits. Previous research mentioned that B. subtilis may cause food spoilage and B. cereus act as food-borne pathogens. They are spore forming bacteria, an important common characteristic is their ability to form endospores that allow them to survive for extended periods under adverse environmental conditions. The aim of this study is to determine antibacterial activity of cardamom against vegetative cell and spores of B.cereus and B. subtilis. In this study, antibacterial activity, in term of MIC and MBC of methanolic extract of cardamom have been tested against B. cereus ATCC33019 and B. subtilis ATCC6633 strains using standard method of Clinical and Laboratory Standard Institute (CLSI). Our result showed that cardamom significantly inhibited the vegetative cells of B. cereus and B.subtilis with minimum inhibitory concentration (MIC) of 1.25 mg/mL. The vegetative cells of B. cereus were completely killed with minimum bactericidal concentration (MBC) of 1.25 mg//mL and B. subtilis with MBC of 10 mg//mL. Antispore activity has been tested against B. cereus and B. subtilis spores of cardamom at concentrations of 0%, 1% and 2% with exposure time of 0 hour and 1 hour. The extract reduced the number of spores from 5.16 to 2.37 of B. cereus, significantly was dependent on concentration. Meanwhile, against B. subtilis was 4.99 To 2.32 and significantly was dependent on exposure incubation time. Overall, methanolic extract of cardamom shows potential antimicrobial and antispore activity against B. cereus and B. subtilis vegetative cells and spores.
