Abstract

Levodopa (L-dopa) merupakan obat untuk mengatasi gangguan motorik penyakit Parkinson, suatu penyakit neurodegeneratif yang disebabkan oleh penurunan sekresi dopamin di otak secara signifikan. Nanoenkapsulasi L-dopa dengan nanoselulosa diharapkan dapat meningkatkan penghantaran L-dopa ke otak. Penelitian ini bertujuan untuk mengetahui kondisi optimum nanoenkapsulasi L-dopa dalam nanoselulosa, serta karakteristik, efisiensi enkapsulasi, dan kemampuan pelepasan obat (drug release) dari produk nanoenkapsulasi tersebut. Nanoenkapsulasi dilakukan dengan metode difusi pelarut yang dimodifikasi pada variasi perbandingan massa pereaksi. Produk nanoenkapsulasi dikarakterisasi menggunakan FTIR, SEM, dan, TEM. Pengujian efisiensi enkapsulasi dan uji drug release dilakukan menggunakan spektofotometer UV-Vis. Hasil penelitian menunjukkan bahwa kondisi optimum nanoenkapsulasi L-dopa dalam nanoselulosa diperoleh pada perbandingan massa L-dopa : nanoselulosa 1:1. Terjadinya nanoenkapsulasi ditunjukkan dari spektrum FTIR melalui adanya pergeseran pada bilangan gelombang 1369,5 cm-1 dari vibrasi NH2 wagging yang menunjukkan terjadi interaksi antara nanoselulosa dengan L-dopa. Berdasarkan hasil karakterisasi SEM, diketahui produk nanoenkapsulasi memiliki bentuk spherical tidak beraturan dengan tekstur permukaan berupa flake (serpihan) yang tidak rata dan belum homogen, sementara itu berdasarkan hasil karakterisasi TEM, diketahui bahwa rentang ukuran partikelnya 207,885-721,254 nm. Berdasarkan pengujian efisiensi enkapsulasi (EE) didapatkan %EE tertinggi sebesar 84,8%. Sementara itu, uji drug release menunjukkan bahwa pada larutan pH 1,2 didapatkan presentase drug release sebesar 5,4% dan pada larutan pH 7,4 didapatkan presentase drug release sebesar 12,3%. Levodopa (L-dopa) is a medicine for Parkinson’s Disease’s motor disorder, a neurodegenerative disease caused by the decrease of dopamine secretion in the brain significally. Nanoencapsulation of L-dopa in nanocellulose is expected to improve the L-dopa delivery to the brain. This study aims to determine the optimum condition for nanoencapsulation of L-dopa in nanocellulose, along with the characteristics, encapsulation efficiency, dan drug relase ability of the nanoencapsulation product. Nanoencapsulation was carried out with a modified solvent diffusion method with a variation in the ratio of reagent mass. Nanoencapsulation products were characterized using FTIR, SEM, and TEM. Encapsulation efficiency and drug release ability test were conducted using UV-Vis spectrophotometer. The results show that the optimum condition for nanoencapsulation of L-dopa in nanocellulose were at the mass ratio of L-dopa : nanocellulose 1:1. The occurrence of encapsulation was shown is indicated by the FTIR spectrum through a shift at wavenumber 1369,5 cm-1 from a vibration of NH2 wagging which shows that there is an interaction between nanocellulose and L-dopa. Based on the SEM characterization results, it was determined that the nanoencapsulation product has an uneven spherical morphology with a surface texture in the form of uneven and not homogeneous. Meanwhile, based on the TEM characterization results, it was determined that the particle size range is between 207,885-721,254 nm. Based on the encapsulation efficiency test (EE), the highest %EE was 84,8%. Meanwhile, the drug release test shows that at pH 1,2 the drug release precentage is 5,4% and at pH 7,4 the drug release percentage is 12,3%.
