Abstract

Andaliman (Zanthoxylum acanthopodium DC.) merupakan tanaman endemik yang tumbuh di Sumatera Utara. Buah andaliman biasa digunakan sebagai salah satu bumbu masakan tradisional khas suku Batak. Tanaman andaliman dilaporkan memiliki banyak aktivitas biologis. Beberapa penelitian melaporkan bahwa tanaman dari genus Zanthoxylum memiliki berbagai kandungan metabolit sekunder yang termasuk golongan flavonoid, lignan, kumarin, terpen, sterol, alkaloid, dan amida. Berdasarkan hal tersebut, penelitian ini bertujuan untuk menambah data kemotaksonomi tanaman Zanthoxylum acanthopodium DC. asal Sumatera Utara. Ekstrak etil asetat buah andaliman dipisahkan dan dimurnikan dengan metode Kromatografi Cair Vakum (KCV) dan Kromatografi Lapis Tipis Preparatif (KLTP) hingga diperoleh senyawa berupa padatan kental berwarna coklat berbau khas sebanyak 71 mg yang diidentifikasi dengan Kromatografi Lapis Tipis (KLT) dengan nilai retardation factor (RF) sebesar 0,65. Struktur senyawa murni ditentukan berdasarkan data spektroskopi proton Nuclear Magnetic Resonance (1H-NMR). Berdasarkan data dari 1H-NMR dan dengan metode membandingkan spektrum 1H-NMR senyawa yang telah diketahui maka diperoleh struktur serta senyawa hasil isolasi ekstrak etil asetat buah andaliman yaitu senyawa ZP-amida F. Kata kunci: Zanthoxylum acanthopodium DC., etil asetat, kromatografi lapis tipis preparatif, ZP-amida F. Andaliman (Zanthoxylum acanthopodium DC.) is an endemic plant that grows in North Sumatra. Andaliman fruit is commonly used as a spice in traditional Batak cuisine. Andaliman plant is reported to have many biological activities. Several studies have reported that plants of the Zanthoxylum genus contain various secondary metabolites, including flavonoids, lignans, coumarins, terpenes, sterols, alkaloids, and amides. Based on this, this study aims to increase the chemotaxonomic data of Zanthoxylum acanthopodium DC. from North Sumatra. The ethyl acetate extract of andaliman fruit was separated and purified using Vacuum Liquid Chromatography (KCV) and Preparative Thin Layer Chromatography (TLC) to obtain a compound in the form of a thick brown solid with a characteristic odor of 71 mg which was identified by Thin Layer Chromatography (TLC) with a retardation factor value. (RF) of 0.65. The structure of the pure compound was determined based on the proton spectroscopic data of Nuclear Magnetic Resonance (1H-NMR). Based on data from 1H-NMR and by comparing the 1H-NMR spectrum of known compounds, the structure and compound isolated from the andaliman fruit ethyl acetate extract, namely the compound ZP-amide F.
