Abstract

ABSTRAK Cyperus rotundus L. atau rumput teki merupakan gulma yang banyak dimanfaatkan dalam bidang medik karena diketahui mengandung berbagai senyawa bioaktif. Tumbuhan ini berasosiasi dengan bakteri endofit yang dapat menghasilkan senyawa bioaktif yang bermanfaat dalam bidang medik. Lambatnya kemajuan dalam penemuan obat antifungi dibandingkan dengan antibakteri menjadi salah satu permasalahan di dunia medik. Tujuan penelitian yaitu untuk mengisolasi dan mengidentifikasi bakteri endofit pada rimpang C. rotundus L. serta mengetahui potensinya sebagai antifungi terhadap C. albicans. Isolasi dilakukan menggunakan metode cawan tuang dan pengenceran. Identifikasi dilakukan dengan mengamati morfologi koloni, reaksi Gram, endospora, serta uji biokimia. Seleksi isolat penghasil senyawa antifungi dilakukan menggunakan metode antagonis. Ekstraksi supernatan dari kultur bakteri terpilih yang ditumbuhkan hingga fase stasioner menggunakan pelarut etil asetat yang kemudian dievaporasi. Digunakan metode Disk Diffusion Assay, Minimum Inhibitory Concentration, dan Minimum Fungicide Concentration untuk pengujian aktivitas antifungi. Hasil penelitian menunjukkan, terdapat 12 isolat bakteri yang teridentifikasi memiliki kemiripan karakteristik dengan genus Bacillus, Neisseria, Shigella, Acinetobacter, Renibacterium, dan Serratia. Uji aktvitas antifungi dilakukan pada isolat A1, A2, dan A4 yang merupakan genus Bacillus terhadap fungi patogen Candida albicans. Ketiga isolat memiliki pengaruh yang signifikan pada konsentrasi 10, 20, 30, 40, dan 50 mg/mL terhadap kontrol positif ketoconazole 30 mg/mL dan kontrol negatif DMSO 10%. Diameter zona hambat tertinggi terdapat pada isolat A4 konsentrasi 50 mg/mL sebesar 21,34±2,5 mm. Nilai MIC dan MFC isolat A1, A2, dan A4 berturut-turut 12.5, 12.5, 6.25 mg/mL dan 25, 25, dan 12.5 mg/mL. Berdasarkan penelitian ini, maka bakteri endofit rimpang C. rotundus memiliki potensi sebagai antifungi. Kata kunci: bakteri endofit, antifungi, Cyperus rotundus, Candida albicans ABSTRACT Purple nutsedge or Cyperus rotundus L. is a wild weed that has been widely used in medical field known to contain bioactive compounds. Purple nutsedge associated with endophytic bacteria that can produce bioactive compounds that are useful in medical field. The discovery of antifungal drugs are relatively slow compared to antibacterial drugs. The aim of this study is to isolate and identify endophytic bacteria in the rhizome of C. rotundus L. and to determine antifungal activities of bacteria against C. albicans. Isolation was conducted using pour plate method and serial dilution. Bacterial are identified by morphology and biochemical test. Bacterial screening carried out using antagonistic assay. The bacteria were grown to stationary phase, supernatant extracted using ethyl acetate, and organic solvent was evaporated. Antifungal activity determinated by Disk Diffusion Assay, Minimum Inhibitory Concentration, and Minimum Fungicide Concentration test. The bacterial isolates were identified as bacterial belonging to Bacillus, Neisseria, Shigella, Acinetobacter, Renibacterium, and Serratia. Antagonistic assay showed that isolates A1, A2, and A4 were able to inhibit the gowth of C. albicans. The isolates had a significant effect at concentrations of 10, 20, 30, 40, and 50 mg/mL on positive control of ketoconazole 30 mg/mL and negative control of DMSO 10%. The highest inhibitory diameter was found in isolate A4 50 mg/mL of 21.34±2.5 mm. MIC and MFC of isolates A1, A2, and A4 are 12.5, 12.5, 6.25 mg/mL, and 25, 25, 12.5 mg/mL respectively.
