Abstract

Hiperkolesterolemia berpotensi pada pembentukan aterosklerosis yang berujung pada PJK (Penyakit Jantung Koroner). Banyak penelitian penurun kolesterol pada pengkondisian hewan menjadi hiperkolesterolemia menggunakan dosis dan durasi pemberian pakan tinggi lemak yang beragam. Kadar kolesterol pada hiperkolesterolemia maupun normal, kurang dikaji terkait jaringan yang terpapar zat peningkat/penurun kolesterol. Dikhawatirkan terjadi perubahan fisiologis jaringan yang mungkin memengaruhi hasil penelitian zat penurun/peningkat kolesterol. Tujuan penelitian ini untuk menginformasikan dosis dan waktu pemberian optimal pada pengkondisian model hewan hiperkolesterolemia yang tidak berbahaya bagi hewan uji. Metode penelitian adalah eksperimental, dengan desain Rancangan Acak Kelompok (RAK) menggunakan kontrol dan perlakuan dosis: Dosis A (1/4 lemak), Dosis B (1/2 lemak), Dosis C (2/3 lemak) dari total pakan maksimal per hari (15 g/ekor/hari) yang diberikan masing-masing selama 1, 2, 3, dan 4 minggu. Pakan berupa campuran pelet kelinci dan gajih sapi. Hewan berupa mencit (Mus musculus) betina usia 3-4 bulan dengan rerata berat 20-25 g sebanyak 48 ekor. Mencit dibunuh, diambil darah dan aorta jantung untuk analisis profil kadar kolesterol dan histologis. Hasil menunjukkan seluruh dosis dapat meningkatkan profil kadar kolesterol. Dosis dan waktu optimal peningkatan profil kadar kolesterol hingga tercapai lesi aterosklerosis awal adalah Dosis B selama 2 minggu. Dosis C tidak aman digunakan karena menyebabkan kondisi tidak sehat pada hewan. Dosis A selama 4 minggu optimal digunakan pada pengkondisian model hewan hiperkolesterolemia yang aman bagi hewan uji. Sementara itu, kadar HDL dapat distabilkan jika terdapat 18% serat dalam pakan standar. Kata Kunci: Hiperkolesterolemia, Aterosklerosis, Model Hewan, Pakan Berlemak, Lemak Sapi. Hypercholesterolemia, due to excessive fat consumption, can form atherosclerosis leading to CHD (coronary heart disease). Many cholesterol-lowering studies in conditioning animals to become hypercholesterolemic considerably apply various doses and durations of the high-fat feed. Cholesterol levels both in hypercholesterolemia and normal have not been examined concerning tissues exposed to cholesterol increasing/lowering substances. It is concerned that there be physiological changes in the tissue that might affect the results of studies of cholesterol-lowering/increasing substances. This study informed the optimal dose and time of administration in conditioning animal models of hypercholesterolemia that are not harmful to test animals. The research method was experimental, with a Complete Randomized Block Design using control and dose treatment: dose A (1/4 fat), dose B (1/2 fat), dose C (2/3 fat) of the maximum total feed per day (15 g/head/day) given for 1, 2, 3, and 4 weeks, respectively. The feed was a mixture of rabbit pellets and beef tallow. Animals in the form of female mice (Mus musculus) aged 3–4 months with an average weight of 20–25 g as many as 48 tails. The mice were killed, blood and aorta were taken for analysis of cholesterol levels and histological profiles. The results indicate that all doses could improve the cholesterol-level profile. Initial atherogenic lesions formed at Dose B for 2 weeks. Dose C is at risk of use because it causes unhealthy conditions in animal models. Dose A for 4 weeks is optimal for use in conditioning animal models of hypercholesterolemia, which is safe for the animals. Additionally, HDL levels can be stabilized if there is 18% fiber in the standard feed. Keywords: Hypercholesterolemia, Atherosclerosis, Animal Model, High Fat diet, Beef Tallow.
