ISOLASI DAN PENENTUAN AKTIVITAS ANTIJAMUR EKSTRAK DAUN SIRSAK ANNONA MURICATA TERHADAP ASPERGILLUS NIGER

    Abstract

    Daun sirsak (Annona muricata) diketahui berfungsi sebagai antimikroba, namun belum banyak penelitian yang diarahkan khusus pada antijamur. Penelitian ini bertujuan untuk mengetahui potensi daun sirsak sebagai antijamur. Penelitian diawali oleh ekstraksi komponen metabolit sekunder daun sirsak menggunakan metode maserasi, dengan 3 jenis pelarut, yaitu metanol, etanol, dan air. Ekstrak yang diperoleh dilakukan pemekatan dan dilanjutkan uji fitokimia. Jamur yang digunakan diisolasi dari bolu kukus, selanjutnya diidentifikasi. Uji Aktivitas antijamur ekstrak daun sirsak menggunakan metode difusi cakram dengan konsentrasi ekstrak, yaitu 3000 ppm, 4000 ppm, 6000 ppm, dan 8000 ppm; serta menggunakan kontrol positif ketokonazol dan kontrol negatif pelarut dari ketiga macam ekstrak. Hasil ekstraksi menunjukkan randemen untuk ekstrak metanol 3,60% (1,82 g); ekstrak etanol 3,64% (1,84 g); dan ekstrak air 2,50% (1,25 g). Hasil uji fitokimia dan pengukuran menggunakan instrumen inframerah diketahui bahwa ekstrak metanol dan etanol memiliki golongan senyawa saponin, tanin, dan steroid, sedangkan ekstrak air hanya memiliki golongan senyawa saponin. Hasil isolasi dan identifikasi jamur pada bolu kukus diketahui bahwa jamur termasuk spesies Aspergillus niger. Hasil penelitian menunjukkan bahwa ekstrak metanol, ekstrak etanol, dan ekstrak air dari daun sirsak dengan konsentrasi 3000 ppm, 4000 ppm, 6000 ppm, dan 8000 ppm diketahui tidak efektif dalam menghambat pertumbuhan Aspergillus Niger.     Soursop leaves (Annona muricata) are known as an antimicrobial agent, but not much research specifically directed to antifungal. This research aims to determine the potential of soursop leaves as an antifungal. Research begins by extracting secondary metabolite components of soursop leaves using maceration method, with 3 types of solvents are methanol, ethanol, and water. Then the extracts are concentrated and continued with phytochemical testing. The fungi were isolated from steamed cake, then identified. Antifungal activity test for soursop leaves extracts using the disc diffusion method with the extracts concentration are 3000 ppm, 4000 ppm, 6000 ppm, and 8000 ppm; and using ketoconazole as positive control and the three kinds of solvent extracts as negative control. Extraction resulted randemen for methanol extract 3.60 % (1.82 g); ethanol extract 3.64 % (1.84 g); and water extract 2.50 % (1.25 g). The results of phytochemical test and measurement using infrared instruments are known that methanol and ethanol extracts have compounds group of saponin, tanin, and steroid, while the water extract contains only compound group of saponin. Isolation and identification of fungi from steamed cake resulted that fungi is included to Aspergillus niger species. Antifungal activity test for each soursop leaves extracts showed that the methanol extract, ethanol extract, and water extract with concentration of 3000 ppm, 4000 ppm, 6000 ppm, and 8000 ppm are not effective to inhibiting the growth of Aspergillus niger.
