Abstract

Produksi minuman ringan kemasan gelas selalu mengalami peningkatan. Minuman ringan kemasan gelas pada umumnya mengandung bahan tambahan pangan seperti pengawet, pemanis, dan pewarna. Bahan tambahan pangan tersebut berpotensi dapat menyebabkan dampak negatif bagi kesehatan manusia. Maka dari itu, dilakukan uji toksisitas akut LC50 – 24 dan 48 jam dengan menggunakan neonate Daphnia magna. Sampel minuman yang diuji toksisitasnya yaitu, TH, NC, TR, dan LT. Minuman-minuman tersebut merupakan hasil seleksi dengan kriteria jumlahnya di agen minuman banyak, tidak terdaftar di Badan Pengawas Obat dan Makanan, tidak tersertifikasi Halal, dan tidak terdapat tanggal kedaluarsa. Hasil uji kandungan siklamat dan benzoat pada keempat sampel minuman gelas, menunjukkan ada kandungan yang melebihi batas kadar maksimum yang sudah ditetapkan. Uji toksisitas ini terdiri dari tahap range finding test dan definitive test dengan menggunakan lima konsentrasi dan kontrol. Neonate dimasukkan kedalam 10 ml sampel minuman, diamati setiap 24 jam dan 48 jam. Medium freshwater digunakan sebagai kontrol dan larutan pengencer sampel. Estimasi nilai LC50-24 jam untuk TH berada pada rentang konsentrasi 30,06-41,15%; NC 17,75-25,44%; TR 9,80-15,54%; dan LT 20,41-25,02%. Sedangkan, untuk nilai LC50-48 jam TH berada pada rentang konsentrasi 22,49-26,01%; NC 16,95-23,87%; TR 7,10-14,22%; dan LT 11,78-21,22%. Keempat minuman bersifat toksik akut bagi neonate D. magna karena nilai LC50-24 jam berada di bawah konsentrasi 42% dan LC50-48 jam berada di bawah konsentrasi 27% serta terjadi dalam waktu yang singkat, yaitu dua hari. ;---The production of beverage in glass packaging is always increasing. Beverages in glass packaging generally contain food additives such as preservatives, sweeteners, and dyes. The food additives potentially cause a negative effect for human health. Therefore, an acute toxicity test of LC50 - 24 and 48 hours by neonate Daphnia magna was carried out. Samples of beverages tested toxicity ie TH, NC, TR, and LT. Those are the result of selection with criteria the number of drinks in the beverage agents are many, not registered in the Food and Drug Administration, not Halal certified, and there are no expired dates. The results of the cyclamate and benzoate test in all samples showed that the chemical subtance exceeded the maximum limit of BPOM and minister of health regulations. This toxicity test consists of the range of finding and definitive tests using five concentrations and controls. Neonate is inserted into 10 ml of beverage sample and observed every 24 hours and 48 hours. Freshwater medium used as a control and dilution solution of the sample. Estimated value of LC50-24 hours for TH is in the concentration range 30,06-41,15%; NC 17,75-25,44%; TR 9,80-15,54%; and LT 20,41-25,02%. Meanwhile, for LC50-48 hours TH is in the concentration range 22,49-26,01%; NC 16,95-23,87%; TR 7,10-14,22%; and LT 11,78-21,22%. The four drinks are acutely toxic to neonate D. magna because LC50-24 hours is <42% and LC50-48 hours is <27%.
