Abstract

Kecombrang (Etlingera elatior) atau honje merupakan salah satu tanaman yang tersebar luas di Indonesia. Tanaman ini biasa digunakan sebagai tanaman obat yang memiliki senyawa antioksidan yang dapat melindungi dan menghentikan reaksi autooksidasi senyawa radikal bebas. Radikal bebas dapat memicu terjadinya penyakit degeneratif akibat penggunaan obat yang memiliki efek genotoksik. Siklofosfamid salah satu obat yang digunakan sebagai antikanker yang memiliki efek imunosupresif, namun memiliki efek samping yang dapat mengakibatkan toksisitas pada gonad. Penelitian ini bertujuan untuk menganalisis pengaruh ekstrak daun kecombrang terhadap kualitas sperma mencit (Mus musculus) jantan yang diinduksi siklofosfamid. Metode penelitian ini berupa Rancangan Acak Lengkap (RAL) dengan 24 ekor yang dibagi menjadi enam kelompok (kontrol negatif, kontrol positif, 5 mg/kg BB, 10 mg/kg BB, 15 mg/kg BB, atau 20 mg/kg BB). Aklimatisasi dilakukan selama tiga minggu, sebelum perlakuan mencit diinduksi siklofosfamid 50 mg/kg BB kemudian diberi perlakuan pemberian ekstrak daun kecombrang selama 14 hari menggunakan jarum gavage. Analisis data statistik menggunakan uji parametik One Way ANOVA. Berdasarkan hasil penelitian pemberian ekstrak daun kecombrang menunjukkan adanya pengaruh signifikan terhadap bobot testis dan tidak berpengaruh signifikan terhadap kualitas sperma (jumlah sperma dan abnormalitas sperma) mencit jantan yang diinduksi siklofosfamid. Adapun dosis ekstrak daun kecombrang yang memiliki nilai paling stabil pengaruhnya terhadap bobot testis yaitu pada dosis 10 mg/kg BB dibandingkan dengan dosis yang lainnya. Penelitian ini menunjukkan bahwa ekstrak daun kecombrang sebagai bahan alami yang memiliki kandungan antioksidan tinggi, namun tidak dapat meningkatkan kualitas sperma mencit jantan.;---Etlingera elatior (kecombrang, honje) is an endemic plant of Indonesia. The plant is used to be utilized as medical herbal, due to its element of anti-oxidant, which functionally known for internal body protection and its potential to stop auto-oxidation attempt from free radical of any chemical exposure. Free radical is known for its potential to trigger effect. Cyclophosphamide is well-known for its usage for anti-cancer, activating immunosuppressive but having side effect to trigger toxic exposure towards the gonad. This research is aimed to analyse the effect of Etlingera elatior leafs extract towards sperm quality of male Mus musculus induced by Cyclophosphamide. The study is run under the methodology of Completely Randomized Design (CRD) subjecting 24 Mus musculus. The subjects are divided into 6 groups (negative control, positive control, 5 mg/kg BW (body weight), 10 mg/kg BW, 15 mg/kg BW and 20 mg/kg BW). The process of acclimatization is done for three weeks. Before the treatment, subject of Mus musculus are induced by 50 mg/kg BW Cyclophosphamide, and then treated by the leaf extract of Etlingera elatior for 14 days by injection using gavage needle. A parametric test through One Way ANOVA is used to analyse the data. The results showed that there was a significant effect on body weight and not significant on sperm quality (number of sperm cells and sperm abnormalities) of male Mus musculus induced by Cyclophosphamide. While the dose leaf extract of Etlingera elatior which has the most stable value of its effect on testicular weight is at a dose of 10 mg/kg body weight compared to the other doses. The research showed that the leaf extract of Etlingera elatior is a natural resource of anti-oxidant, but cannot improve the quality of male mice sperm.
