Abstract

Penggunaan tanaman sebagai bahan baku pembuatan obat modern maupun obat tradisional mengalami peningkatan. Salah satu tanaman yang memiliki potensi sebagai bahan obat-obatan adalah Coix lacryma jobi L. (hanjeli). Produksi senyawa metabolit sekunder dari tumbuhan dengan skala yang besar dapat dihasilkan melalui teknik in vitro. Penelitian ini bertujuan untuk menganalisis respons eksplan daun C. lacryma-jobi L. dan untuk ditemukannya rentang konsentrasi yang yang paling optimal untuk menginduksi kalus. Eksplan yang digunakan adalah helaian dan pelepah daun. Eksplan dikultur pada medium N6 dengan penambahan ZPT 2,4-D dengan konsentrasi 0 ppm, 4 ppm, 4,5 ppm, 5 ppm, 5,5 ppm. Kultur dikultivasi selama 8 minggu dalam keadaan gelap. Hasil menunjukan eksplan helaian daun setelah dikultur selama 8 minggu tidak menunjukkan respons. Eksplan pelepah daun menunjukan respons pembesaran jaringan pada 0 ppm (2,67%), 4 ppm (12%), 4,5 ppm (8%), 5 ppm (4%), 5,5 ppm (0%) dan pembentukan kalus pada 0 ppm (0%), 4 ppm (4%), 4,5 ppm (0%), 5 ppm (5,33%), dan 5,5 ppm (0%). Pada eksplan pelepah daun memberikan dua respon yaitu pembesaran jaringan dan pembentukan kalus,sedangkan helaian daun tidak memberikan respons apapun. Pembesaran jaringan tertinggi (12%) didapatkan dari konsentrasi 2,4-D 4 ppm dan pembentukan kalus tertinggi (5,33%) didapatkan dari konsentrasi 2,4-D 5 ppm. One of the plants that have potential to be used as medicine is Coix lacryma jobi L. (Job tears). A large scale production of second metabolite compound can be dobe by in vitro technique. This study is aimed to analyze response of the Coix lacryma jobi L. leaf explants and to find the most optimal range of concentration for induce callus. Explants used were leaf and midrib. The explants were cultured in N6 medium with plant growth regulators (PRG) 2,4-D with concentrations of 0 ppm, 4 ppm, 4,5 ppm, 5 ppm, 5,5 ppm. The cultures were cultivated for 8 weeks in dark condition. The leaf explants cultured in the N6 medium for 8 weeks did not showed any response, meanwhile for midrib explants showed enlargement of tissue percentage, as 0 ppm (2,67%), 4 ppm (12%), 4,5 ppm (8%), 5 ppm (4%), 5,5 ppm (0%) and percentage for callus formation, as 0 ppm (0%), 4 ppm (4%), 4,5 ppm (0%), 5 ppm (5,33%), and 5,5 ppm (0%). In leaf midrib explant, provide two responses, namely tissue enlargement and callus formation, while leaf blade do not provide any response. The highest percentage of tissue enlargement was (12%) which obtained from concentrations of 2,4-D 4 ppm and the highest percentage of callus formation was (5,33%) which obtained from concentrations of 2,4-D 5 ppm.
