Abstract

Pisang menjadi salah satu komoditas buah–buahan yang saat ini cukup diperhitungkan. Semua bagian tanaman pisang dapat digunakan untuk makanan, makanan ternak, serat, tanaman hias dan lain - lain. Penelitian untuk mengevaluasi hubungan filogenetik kultivar pisang (Musa spp.) di Bali berdasarkan sekuen DNA daerah Internal Transcribed Spacer (ITS) - 2 telah dilakukan. Sebanyak 27 sekuen ITS kultivar pisang dan dua sekuen ITS dari jenis Ensete yang berasal dari GeneBank sebagai outgroup digunakan untuk analisis. DNA tanaman diekstraksi dengan metode CTAB dengan sedikit modifikasi. Primer ITS-5 dan ITS-4 digunakan untuk mengamplifikasi sekuen daerah ITS. Data hasil sekuensing digabungkan menggunakan CodonCode Aligner dan disejajarkan menggunakan ClustalX. Pohon filogenetik dibangun menggunakan PAUP*4.0b10. Analisis filogenetik menggunakan metode maximum parsimony berhasil merekonstruksi tiga kelompok monofiletik besar dengan nilai bootstrap yang baik (>50%), tetapi ada enam kultivar terpisah dari tiga kelompok yang terbentuk pada pohon filogenetik. Secara umum, kajian ini menyediakan informasi dasar untuk hibridisasi yang akan membantu meningkatkan nilai ekonomi tanaman pisang. ----- Bananas become one of the fruit commodity which is currently quite accounted. All parts of plant can be used for food, fodder, fiber, ornamental plants and others. A study to evaluate phylogenetic relationship of banana cultivars (Musa spp.) in Bali based on DNA sequences of Internal Transcribed Spacer (ITS)-2 region has been conducted. A total of 27 DNA sequences of ITS region of banana cultivars and two outgroup sequences of the Ensete species form GenBank were used for analysis. DNA was extracted by CTAB method with slight modification. ITS-5 and ITS-4 primers were used to amplify the ITS region sequences. Data from DNA sequencing was contiged using CodonCode Aligner and was aligned using ClustalX. The phylogenetic tree was reconstructed using PAUP*4.0b10. Maxiumum parsimony analysis revealed three major monophyletic groups with good bootstrap value (>50%), but there are six cultivars are separated position from the three groups on the phylogenetics trees. Overall, this study provides a basic information for hybridization in bananas which will help increasing their economic value.
