Abstract

Temulawak (Curcuma xanthorrhiza Roxb.) adalah salah satu tumbuhan obat keluarga Zingiberaceae yang biasa digunakan sebagai bahan baku obat tradisional di Indonesia. Komponen aktif dalam rimpang temulawak yaitu senyawa fenol dapat mengurangi kerusakan oksidatif pada sel akibat dari radikal bebas. Penelitian ini bertujuan untuk menganalisis aktivitas antioksidan dan antihemolisis pada rimpang temulawak. Bahan baku yang digunakan adalah rimpang temulawak berusia 7-9 bulan. Metode ekstraksi menggunakan ekstraksi maserasi dengan melakukan perendaman menggunakan pelarut etanol 95%. Ekstrak rimpang temulawak dianalisis kandungan total fenol. Pengujian antioksidan dilakukan menggunakan metode DPPH (2,2-difenil-1-pikrilhidrazil) dan pengujian antihemolisis dilakukan menggunakan eritrosit manusia dengan pemberian hidrogen peroksida (H2O2). Data yang diperoleh diolah dan dianalisis dengan menggunakan program Microsoft Excel dan SPSS 21. Hasil penelitian pada ekstrak rimpang temulawak ditemukan kandungan total fenol sebesar 120,097 mgGAE/100g sampel. Adanya perbedaan konsentrasi pada ekstrak rimpang temulawak terhadap aktivitas antioksidan dan antihemolisis memengaruhi proses penghambatan radikal bebas DPPH dan stress oksidatif dari H2O2. Nilai IC50 (Inhibition Concentration) aktivitas antioksidan ekstrak rimpang temulawak sebesar 86,350 ppm tergolong aktif dalam menangkap radikal DPPH. Nilai EC50 (Effective Concentration) aktivitas antihemolisis sebesar 57,9293 ppm tergolong aktif dalam menghambat hemolisis pada eritrosit. Korelasi kadar total fenol dengan aktivitas antioksidan sebesar 0,920 dan korelasi kadar total fenol dengan aktivitas antihemolisis sebesar 0,846 menunjukkan adanya hubungan yang kuat terhadap aktivitas antioksidan dan antihemolisis. Ekstrak rimpang temulawak memiliki aktivitas antioksidan dan antihemolisis yang tergolong aktif sehingga berpotensi sebagai antioksidan dan antihemolisis yang baik.----Temulawak (Curcuma xanthorrhiza Roxb.) is the one of Zingiberaceae family of medicinal plants, which commonly used as raw material of traditional medicine in Indonesia. The active component in the temulawak rhizome is phenol compounds can reduce oxidative damage to cells resulting from free radicals. This study aims to analyze the antioxidant activity and antihemolytic on the temulawak rhizome. The raw materials used are 7-9 months old of temulawak’s rhizome. The extraction method uses maceration extraction by immersion using 95% ethanol solvent. The extract of temulawak rhizome was analyzed to calculate total phenol content. Antioxidant was tested by using DPPH method (2,2-diphenyl-1-picrylhydrazyl) and the antihemolytic was tested by using human erythrocytes with the hydrolyzed of hydrogen peroxide (H2O2). The data obtained was processed and analyzed by using the Ms. Excel and SPSS 21 programs. The results of the research on the extract of temulawak rhizome found total phenol content of 120.097 mgGAE / 100g sample. The differences of concentration of temulawak rhizome extract to antioxidant activity and antihemolytic influence the inhibition process of free radical of DPPH and oxidative stress from H2O2. IC50 (Inhibition Concentration) value of the antioxidant activity of temulawak rhizome extract of 86,350 ppm classified as active in capturing DPPH radical. The EC50 (Effective Concentration) value of the antihemolytic activity of 57,9293 ppm is classified as active in inhibiting hemolysis in erythrocytes. The correlation between the levels of total phenol with the antioxidant activity of 0.920 and correlation between levels of total phenol with the antihemolytic activity of 0.846 showed a strong relationship to antioxidant activity and antihemolytic. Extracts of temulawak rhizome have antioxidant activity and antihemolytic which is classified as active, thus potency as good antioxidant and antihemolytic.
