Abstract

Karabenguk (Mucuna pruriens Lin var. Utilis) telah banyak digunakan dalam penanganan penyakit Parkinson. Pengubahan ukuran partikel ekstrak biji karabenguk menjadi nanopartikel diharapkan dapat meningkatkan aktivitas antiparkinsonnya. Tujuan dari penelitian ini untuk melakukan sintesis dan karakterisasi perak-ekstrak biji karabenguk-nanopartikel (AgMPn) dan uji aktivitas antiparkinsonnya pada mencit. Tahapan penelitian yang dilakukan terdiri dari ekstraksi serbuk biji karabenguk, sintesis dan karaterisasi AgMPn, dan uji aktivitas antiparkinson dari AgMPn. Ekstraksi serbuk biji karabenguk dilakukan dengan metode maserasi menggunakan pelarut etanol-air (1:1) pada pH 3. Sintesis AgMPn dilakukan dengan mereaksikan larutan perak nitrat dan ekstrak biji karabenguk dengan perbandingan massa 1:5 selama 40 menit. Karakterisasi AgMPn dilakukan menggunakan instrumen Fourier Transform Infrared (FTIR), Transmission Electron Microscopy (TEM), dan Scanning Electron Microscopy-Energy Dispersive X-Ray (SEM-EDX). Uji aktivitas antiparkinson dari AgMPn dilakukan melalui uji katalepsi pada mencit yang diinduksi dengan haloperidol. Uji katalepsi dilakukan pada dosis 5, 10, 15, 20, dan 25 mg/kg berat badan mencit. Hasil ekstraksi serbuk biji karabenguk diperoleh ekstrak biji karabenguk kering sebanyak 4,11%. Hasil sintesis AgMPn diperoleh serbuk berwarna hitam sebanyak 5,2%. Berdasarkan hasil karakterisasi menggunakan TEM dan SEM-EDX, diketahui bahwa AgMPn hasil sintesis berbentuk bulat (spherical) dengan ukuran partikel 21-117 nm dan mengandung unsur perak (Ag), karbon (C), oksigen (O), dan nitrogen (N). Spektrum FTIR dari AgMPn menunjukkan pita serapan IR pada daerah 1512 cm-1 yang mengindikasikan adanya interaksi antara logam perak dengan atom N pada AgMPn. Hasil uji aktivitas antiparkinson pada mencit menunjukkan AgMPn dapat menurunkan gejala katalepsi, dan dari kelima dosis yang diuji, dosis yang paling baik dalam menurunkan gejala katalepsi adalah dosis 5 mg/kg berat badan.---------- Velvet bean (Mucuna pruriens Linn var. Utilis) has been used for Parkinson disease’s treatment. Transforming the particle size of velvet bean extract into nanoparticles was expected to increase its antiparkinson activity. The purpose of this research was to synthesis and characterize the silver-velvet bean extract- nanoparticles (AgMPn) and to evaluate its antiparkinson activity in mice. The research consisted of three stages, which are. extraction of velvet bean powder, synthesis and characterization of AgMPn, and evaluation of antiparkinson activity of AgMPn. The extraction of velvet bean powder was done by maceration using ethanol-water (1: 1) at pH 3. The synthesis of AgMPn was done by reacting the silver nitrate solution and the velvet bean extract with mass ratio of 1:5 for 40 minutes. Characterization of synthesis product was performed using fourier transform infrared (FTIR), transmission electron microscopy (TEM), and scanning electron microscopy-energy dispersive X-ray (SEM-EDX) instruments. The evaluation of antiparkinson activity of AgMPn was performed through a cataleptic test in haloperidol-induced mice. The catalepsy test was performed at several dosage, i.e. 5, 10, 15, 20, and 25 mg/kg body weight. The extraction of velvet bean powder produced a dry extract in 4.11%. AgMPn obtained from synthesis was a powder with 5.2% of rendement. Based on the characterization using TEM and SEM-EDX, AgMPn contains silver (Ag), carbon (C), oxygen (O), and nitrogen (N) with spherical in shape and has particle size of 21-117 nm. The FTIR spectra of AgMPn showed absorption bands at 1512 cm-1 indicated interaction between silver with nitrogen site of AgMPn. The results of antiparkinson test in mice showed AgMPn could reduced the catalepsy symptoms, and the best dosage was 5 mg/kg body weight.
