Abstract

Telah dilakukan penelitian tentang pengaruh ekstrak rimpang jahe merah (Zingiber officinale var. Rubrum) terhadap perkembangan embrio praimplantasi mencit (Mus musculus) Swiss Webster. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak rimpang jahe merah terhadap tahapan perkembangan embrio praimplantasi, abnormalitas embrio praimplantasi, dan ukuran diameter blastokista. Mencit betina yang digunakan berjumlah 24 ekor yang secara acak dibagi menjadi empat kelompok yaitu kelompok kontrol dan tiga kelompok perlakuan dengan pengulangan sebanyak 6 kali. Pemberian ekstrak jahe merah diberikan secara gavage dengan konsentrasi 0 mg/kgBB/hari, 140 mg/kgBB/hari, 280 mg/kgBB/hari, dan 700 mg/kgBB/hari setelah ditemukannya vaginal plug. Pada hari kebuntingan ke-3,5 mencit didislokasi leher kemudian organ reproduksi diisolasi. Embrio dikeluarkan dari uterus dengan teknik flushing kemudian dihitung jumlahnya dan dianalisis. Berdasarkan uji statistik, seluruh dosis perlakuan ekstrak rimpang jahe merah hanya berpengaruh secara signifikan pada blastokista. Ketiga dosis perlakuan tidak memiliki perbedaan nyata namun efek menghambat perkembangan embrio praimplantasi sudah terlihat dari dosis terendah yaitu 140 mg/kgBB/hari jika dibandingkan dengan kelompok kontrol. Persentase terbentuknya embrio praimplantasi abnormal semakin meningkat jika dibandingkan dengan kelompok kontrol. Selain itu, uji statistika menunjukkan bahwa ekstrak rimpang jahe merah berpengaruh secara signifikan terhadap ukuran diameter blastokista. Hasil dari penelitian ini menunjukkan bahwa ekstrak rimpang jahe merah dapat menghambat perkembangan embrio praimplantasi mencit. Kata Kunci: Research on the effect of red ginger (Zingiber officinale var. Rubrum) rhizome on embryo preimplantation development of mice (Mus musculus) swiss webter has done. The purpose of this study was to determine the effect of red ginger rhizome on embryo preimplantation stages, abnormality of embryo preimplantation, and diameter of blastocyst. Female mice were used each treatment totaling 24 tails were divided randomly into control group and three treatment groups. Mice were administered weith red ginger extract dose 0 mg/bw/day, 140 mg/bw/day, 280 mg/bw/day, and 700 mg/bw/day by gavage after a vaginal plug detected. On gestion day 3.5 mice were sacrificed and the reproductive organ were isolated. Embryos were collected by flushing for counted and analyzed. Based on statistical test, whole of treatment doses only showed significantly (p<0,05) effect to blastocysts. Red ginger extract could decrease the percentage of embryos that reached blastocysts, this was caused by increase of the percentage of embryos that could only reach the morula stage. Percentage of embryo abnormal were increase compared with control group. Beside that, statistical test showed that red ginger extract coul decrease significantly (p<0,05) the diameter of blastocyst. It could be conclude that the administration of ;red ginger rhizome extract on gestation day 0 inhibit the development of mice preimplantation embryos. Keywords: Zingiber officinale var.
