Abstract
Rimpang kunyit (Curcuma domestica Val.) mengandung beberapa komponen kimia yang mampu menyebabkan terhambatnya pembelahan sel. Penelitian ini bertujuan untuk melihat pengaruh pemberian ekstrak rimpang kunyit terhadap perkembangan embrio praimplantasi mencit (Mus musculus) Swiss Webster. Mencit diaklimatisasi selama tujuh hari lalu dikawinkan dan setelah adanya vaginal plug mencit diberi perlakuan ekstrak rimpang kunyit. Dosis kunyit yang digunakan adalah 0 mg/kg bb/hari, 140 mg/kg bb/hari, 280 mg/kg bb/hari, dan 700 mg/kg bb/hari. Pemberian ekstrak dilakukan secara gavage selama mencit bunting dari hari ke-0 sampai hari ke-3. Pengamatan dilakukan terhadap hasil dari koleksi embrio dengan teknik flushing menggunakan medium PBS (Phosfat Buffer Saline). Pada tahapan ini dilihat persentase tahapan perkembangan embrio, abnormalitas embrio dan ukuran diameter embrio blastokista. Hasil penelitian menunjukkan bahwa adanya penurunan jumlah embrio yang mencapai tahap blastokista sejalan dengan naiknya dosis yang diberikan dibandingkan kontrol. Selain itu, ada peningkatan pada jumlah embrio tahap morula dan embrio abnormal yang ditemukan sejalan dengan naiknya dosis yang diberikan dibandingkan kontrol. Walaupun terdapat penurunan persentase embrio blastokista, namun secara statistik hasil tersebut tidak meunjukkan hasil yang berbeda secara signifikan. Pengamatan dilakukan pula terhadap ukuran diameter embrio blastokista dan menunjukkan hasil bahwa ukuran diameter blastokista pada kelompok kontrol dan perlakuan tidak jauh berbeda yaitu dengan rata-rata 0,09 ± 0,008 untuk diameter horizontal dan 0,09 ± 0,005 untuk diameter vertikal. Jadi dapat disimpulkan bahwa pemberian ekstrak rimpang kunyit pada mencit yang bunting selama 3,5 hari berpengaruh terhadap perkembangan embrio praimplantasi walaupun secara statistik tidak berbeda secara signifikan. The aim of this research is to know the influence of giving tumeric extract on preimplantation embryo development of mice. Mice were acclimatized for 7 days and then mated. If the vaginal plug is found, then it will be treated by turmeric extract. The dose of the extract were 0 mg / kg bw / day, 140 mg / kg bw / day, 280 mg / kg bw / day, and 700 mg / kg bw / day. The extract was done by gavage for pregnant mice from 0 upto 3.5 days. The observations to result of embryo collections were done by flushing technic used PBS (Phosphate Buffer Saline) medium. At this step, the percentage of embryo development (the abnormality of embrio and the diameter size of blastocysta embryo) were seen. The result of this research showed that there was the decrease of total blastocysta embryo as long as the increase of dose that was given, if it compared with the control. Beside that, there was the decrease of total morula embryo and the abnormality as long as the increase of dose that was given, if it compared with the control. Although, there were the increase in the percentage of embryos, but statistically the results did not show significantly different. In the embryo diameter, obtained results were not much different from the control. It concluded that the rhizoma of tumeric extract, which was performed by gevage to pregnant mice for 3,5 days influencing to preimplantation embryo development, even though in statistic there were not significantly different.
