    Abstract

    Minat terhadap tanaman Dendrobium sebagai tanaman hias dan bunga potong terus meningkat. Namun, budidaya Dendrobium secara konvensional berlangsung lambat. Oleh karena itu, perlu alternatif budidaya menggunakan kultur jaringan. Tujuan dari penelitian ini adalah untuk mendapatkan kombinasi dan konsentrasi BAP, NAA, dan ekstrak tomat terbaik dalam menginduksi tunas aksilar pada batang anggrek Dendrobium sonia pada medium ½ Murashige-Skoog. Penelitian dilakukan dengan menggunakan Rancangan Acak Lengkap (RAL) faktorial dengan 3 faktor yaitu, BAP (2 ppm dan 3 ppm), NAA (0,5 ppm dan 1 ppm), dan ekstrak tomat (5%, 10%, dan 15%). Prosedur penelitian yang dilakukan berupa persiapan penelitian, sterilisasi dan penanaman eksplan, serta pengamatan dan pengambilan data. Hasil penelitian menunjukkan respons eksplan nodus Dendrobium sonia yang dikultur berupa tiga jenis respons, yaitu eksplan tunas terinduksi, eksplan yang bertahan hijau, dan eksplan yang mengalami browning. Eksplan tunas terinduksi muncul dikombinasi 3 ppm BAP + 15% ekstrak tomat; 3 ppm BAP + 0,5 ppm NAA + 10% ekstrak tomat; 3 ppm BAP + 0,5 ppm NAA + 15% ekstrak tomat, dan 3 ppm BAP + 1 ppm NAA + 15% ekstrak tomat. Respons eksplan bertahan hijau dan browning terjadi pada semua kombinasi perlakuan. Berdasarkan penelitian ini disimpulkan bahwa pada medium ½ MS dengan penambahan kombinasi 3 ppm BAP + 0,5 ppm NAA + 10% ekstrak tomat merupakan perlakuan paling baik karena mampu menginduksi tunas aksilar dengan persentase tertinggi yaitu 50%, mempercepat waktu kemunculan tunas yaitu pada minggu ke-3 setelah penanaman, serta dapat mempertahankan warna hijau dengan persentase tertinggi sebesar 80%. Interest in Dendrobium plants as ornamental plants and cut flowers continues to increase. However, conventional Dendrobium cultivation is slow. Therefore, alternative cultivation using tissue culture is needed. The purpose of this study was to obtain the best combination and concentration of BAP, NAA, and tomato extract in inducing axillary shoots in Dendrobium sonia orchid stems on ½ Murashige-Skoog medium. The study was conducted using a factorial Completely Randomized Design (CRD) with 3 factors, namely, BAP (2 ppm and 3 ppm), NAA (0.5 ppm and 1 ppm), and tomato extract (5%, 10%, and 15%). The research procedures carried out included research preparation, sterilization and planting of explants, as well as observation and data collection. The results showed that the responses of cultured Dendrobium sonia nodal explants were in three types of responses, namely induced shoot explants, explants that remained green, and explants that experienced browning. Induced shoot explants emerged in the combinations of 3 ppm BAP + 15% tomato extract; 3 ppm BAP + 0.5 ppm NAA + 10% tomato extract; 3 ppm BAP + 0.5 ppm NAA + 15% tomato extract, and 3 ppm BAP + 1 ppm NAA + 15% tomato extract. Explant responses remained green and browning occurred in all treatment combinations. Based on this study, it was concluded that the ½ MS medium with the addition of a combination of 3 ppm BAP + 0.5 ppm NAA + 10% tomato extract was the best treatment because it was able to induce axillary shoots with the highest percentage of 50%, accelerated the time of shoot emergence to the 3rd week after planting, and could maintain the green color with the highest percentage of 80%.
