    Abstract

    Cabe jawa (Piper retrofractum) merupakan tanaman obat tradisional yang diketahui mengandung senyawa aktif piperin, sejenis alkaloid dengan berbagai aktivitas farmakologis. Penelitian ini bertujuan untuk mengisolasi dan mengkarakterisasi senyawa piperin dari buah cabe jawa, serta mengevaluasi potensinya sebagai inhibitor enzim asetilkolinesterase (AChE) melalui pendekatan in silico. Proses isolasi dilakukan dengan metode maserasi menggunakan pelarut etil asetat, diikuti fraksinasi cair–cair dan pemisahan menggunakan Kromatografi Cair Vakum (KCV) serta Kromatografi Lapis Tipis (KLT). Senyawa hasil isolasi kemudian dikarakterisasi menggunakan spektroskopi UV-Vis, 1H-NMR, 13C-NMR, HSQC, dan HMBC. Hasil analisis menunjukkan bahwa struktur senyawa yang diperoleh sesuai dengan karakteristik piperin berdasarkan data spektroskopi dan literatur pembanding. Uji molecular docking terhadap enzim AChE (PDB ID: 4EY7) menunjukkan bahwa piperin memiliki afinitas ikatan sebesar –10,1 kcal/mol dan mampu berinteraksi dengan residu-residu kunci enzim, seperti TRP 286 dan TYR 337, serupa dengan ligan pembanding donepezil. Berdasarkan hasil tersebut, senyawa piperin dari buah cabe jawa berpotensi sebagai kandidat inhibitor AChE. Javanese chili (Piper retrofractum) is a traditional medicinal plant known to contain the active compound piperine, a type of alkaloid with various pharmacological activities. This study aimed to isolate and characterize piperine from Javanese chili fruit and to disseminate its potential as an acetylcholinesterase (AChE) inhibitor through an in silico approach. The isolation process was carried out using a maceration method using ethyl acetate as a solvent, followed by liquid-liquid fractionation and labeling using Vacuum Liquid Chromatography (VLC) and Thin Layer Chromatography (TLC). The isolated compounds were then characterized using UV-Vis spectroscopy, H-NMR, C-NMR, HSQC, and HMBC. The analysis results showed that the structure of the obtained compounds matched the characteristics of piperine based on spectroscopic data and literature comparisons. Molecular docking of the AChE enzyme (PDB ID: 4EY7) showed that piperine has a binding affinity of –10.1 kcal/mol and is able to interact with key residues of the enzyme, such as TRP 286 and TYR 337, similar to the reference ligand donepezil. Based on these results, the piperine compound from Javanese chili fruit has potential as a candidate AChE inhibitor.
