Abstract
Mikroalga diketahui mampu menghasilkan berbagai senyawa bioaktif bernilai tinggi, tetapi profil biokimianya sangat dipengaruhi oleh medium kultur yang digunakan. Variasi nutrisi dan sumber karbon dalam medium dapat mengarahkan jalur metabolisme sehingga memengaruhi prioritas akumulasi metabolit sel. Dalam penelitian ini, dilakukan analisis pengaruh variasi medium kultur terhadap profil biokimia dan aktivitas antioksidan ekstrak etanol Chlorella sorokiniana wild type dan mutan. Hasil penelitian menunjukkan bahwa medium GYP memberikan hasil terbaik pada beberapa parameter, yaitu menghasilkan biomassa dan kandungan karbohidrat tertinggi, total fenolik paling besar, serta akumulasi pigmen (klorofil dan karotenoid) yang lebih tinggi. Selain itu, aktivitas antioksidan tertinggi juga diperoleh pada medium GYP, ditunjukkan dengan nilai IC₅₀ terendah, khususnya pada galur mutan. Sementara itu, medium TAP lebih berperan dalam meningkatkan akumulasi protein. Temuan ini menegaskan bahwa pemilihan medium kultur sangat menentukan produksi metabolit primer dan sekunder, di mana medium GYP berpotensi lebih optimal untuk meningkatkan biomassa, kandungan senyawa bioaktif, serta memperkuat bioaktivitas ekstrak Chlorella sorokiniana. Kata Kunci: Chlorella sorokiniana, variasi medium, upstream, downstream Microalgae are known to produce a variety of high-value bioactive compounds, but their biochemical profiles are strongly influenced by the culture medium used. Variations in nutrients and carbon sources within the medium can direct metabolic pathways, thereby affecting the priority of metabolite accumulation in cells. In this study, the effects of different culture media on the biochemical profile and antioxidant activity of ethanol extracts from wild-type and mutant Chlorella sorokiniana were analyzed. The results showed that the GYP medium provided the best outcomes in several parameters, including the highest biomass and carbohydrate content, the greatest total phenolic content, as well as higher accumulation of pigments (chlorophyll and carotenoids). In addition, the highest antioxidant activity was also obtained in the GYP medium, as indicated by the lowest IC₅₀ value, particularly in the mutant strain. Meanwhile, the TAP medium played a greater role in enhancing protein accumulation. These findings confirm that the choice of culture medium is crucial for determining the production of primary and secondary metabolites, with the GYP medium being more optimal for enhancing biomass, bioactive compound content, and strengthening the bioactivity of Chlorella sorokiniana extracts.
