Abstract
Tanaman Genus Taxus telah dikenal memiliki potensi antikanker sejak tahun 1972 yang pertama kali diidentifikasi dari ekstrak Taxus brevifolia Nutt di Amerika Serikat. Di Indonesia, satu-satunya spesies yang termasuk Genus ini adalah Taxus sumatrana. Penelitian terdahulu menemukan senyawa antikanker pada kulit batang dan batang muda. Penelitian ini bertujuan untuk mengidentifikasi kandungan metabolit dan aktivitas antioksidan pada dua tingkat perkembangan daun T. sumatrana, yaitu daun dewasa dan daun tua. Daun diambil dari Kebun Raya Cibodas, Kabupaten Cianjur, dari satu bagian batang yang sama pada lima individu berdasarkan keseragaman morfologi. Daun dikeringkan, dihaluskan, dan diekstrak dengan metode maserasi menggunakan pelarut etanol 70% p.a. Kandungan metabolit pada daun dianalisis menggunakan instrumen Gas Chromatography-Mass Spectrometry (GC-MS) dan diidentifikasi berdasarkan indeks kesamaannya dengan pustaka National Institute of Standards Technology (NIST))dan PubChem. Aktivitas antioksidan dianalisis menggunakan metode DPPH dengan pelarut metanol menggunakan spektrometer pada panjang gelombang 515 nm. Hasil penelitian menunjukkan daun dewasa mengandung 9 senyawa, sedangkan daun tua mengandung 13 senyawa. Kedua tingkat daun mengandung 4 senyawa yang sama dan didominasi oleh senyawa metil palmitat. Hasil uji aktivitas antioksidan pada daun dewasa sebesar 13,82 ppm dan daun tua sebesar 20,95 ppm yang termasuk dalam kategori sangat kuat. Penelitian ini menunjukkan adanya perbedaan kandungan metabolit pada tingkat perkembangan daun T. sumatrana, dan aktivitas antioksidan ekstrak daun dewasa lebih tinggi dibanding ekstrak daun tua. The Taxus genus has been recognized for its anticancer potential since 1972, first identified from Taxus brevifolia Nutt extract in the United States. In Indonesia, the only species belonging to this genus is Taxus sumatrana. Previous studies have found anticancer compounds in the bark and young stems. This study aims to identify the metabolite content and antioxidant activity in two developmental stages of T. sumatrana leaves, namely mature and senescent leaves. Leaves were collected from the Cibodas Botanical Garden, Cianjur Regency, from the same branch section on five individuals selected for morphological uniformity. The leaves were dried, ground, and extracted by maceration using 70% ethanol (p.a.) as the solvent. Metabolite contents were analyzed using Gas Chromatography-Mass Spectrometry (GC-MS) and identified based on their similarity index with the National Institute of Standards and Technology (NIST) and PubChem libraries. Antioxidant activity was analyzed using the DPPH method with methanol as a solvent and measured by spectrophotometry at a wavelength of 515 nm. The results showed that mature leaves contained 9 compounds, while senescent leaves contained 13 compounds. Both leaf stages shared 4 common compounds and were dominated by methyl palmitate. Antioxidant activity tests showed values of 13.82 ppm for mature leaves and 20.95 ppm for senescent leaves, both classified as very strong. This study demonstrates differences in metabolite content across T. sumatrana leaf development stages, with mature leaf extracts exhibiting higher antioxidant activity than senescent leaf extracts.
