Abstract

Kariofilena merupakan seskuiterpena yang banyak ditemukan dalam minyak esensial dan dikenal memiliki berbagai aktivitas biologis yang potensial. Namun, pendayagunaan dan sintesis kariofilena beserta turunannya masih minim diteliti di Indonesia. Sintesis turunan kariofilena sering kali menemui tantangan, seperti rendahnya selektivitas dan terbentuknya produk samping yang tidak diinginkan. Sebelumnya, percobaan menggunakan oksigen sebagai agen pengoksidasi telah menunjukkan hasil yang menjanjikan tetapi terbentuknya produk samping masih menjadi kendala utama. Ozon, sebagai oksidator yang lebih kuat dibandingkan oksigen, dipilih sebagai agen pengoksidasi alternatif karena struktur molekulnya yang lebih reaktif. Penelitian ini bertujuan untuk mengidentifikasi produk reaksi kariofilena dengan ozon serta menentukan kondisi optimum untuk sintesis senyawa turunan kariofilena menggunakan ozon. Reaksi dilakukan dalam tabung reaktor dengan bantuan generator ozon. Variasi kondisi terdiri atas variasi suhu pada 50oC, 60oC, 70oC, dan 80oC serta variasi waktu selama 6 jam, 7 jam, dan 8 jam. Produk reaksi dianalisis menggunakan Gas Chromatography-Flame Ionization Detector (GC-FID) dan Gas Chromatography-Mass Spectrometry (GC-MS). Hasil penelitian menunjukkan bahwa reaksi antara kariofilena dan ozon menghasilkan senyawa kariofilena oksida. Kondisi optimal dicapai pada suhu 80°C dan waktu reaksi 7 jam. Analisis GC-FID menghasilkan konversi produk sebesar 66,654% pada waktu retensi 20,055 tanpa terjadinya reaksi samping. Hasil uji GC-MS menunjukkan konversi produk sebesar 54,58% pada waktu retensi 17,979, disertai dengan adanya campuran pengotor. Caryophyllene is a sesquiterpene found in many essential oils and is known to have various potential biological activities. However, the utilization and synthesis of caryophyllene and its derivatives have been minimally studied in Indonesia. The synthesis of caryophyllene derivatives often encounters challenges, such as low selectivity and the formation of unwanted by-products. Previously, experiments using oxygen as an oxidizing agent have shown promising results but the formation of by-products is still a major obstacle. Ozone, as a stronger oxidizer than oxygen, was chosen as an alternative oxidizing agent because of its more reactive molecular structure. This study aims to identify the reaction products of caryophyllene with ozone and determine the optimum conditions for the synthesis of caryophyllene derivative compounds using ozone. The reaction was carried out in a reactor tube with the help of an ozone generator. The variation of conditions consisted of temperature variations of 50°C, 60°C, 70°C, and 80°C and time variations of 6 hours, 7 hours, and 8 hours. The reaction products were analyzed using Gas Chromatography-Flame Ionization Detector (GC-FID) and Gas Chromatography-Mass Spectrometry (GC-MS). The results showed that the reaction between caryophyllene and ozone produces caryophyllene oxide. The optimal conditions were achieved at a temperature of 80°C and a reaction time of 7 hours. GC-FID analysis yielded a product conversion of 66.654% at a retention time of 20.055, without any side reactions. The GC-MS test results showed a product conversion of 54.58% at a retention time of 17.979, accompanied by the presence of impurities.
