Abstract
Penelitian mengenai pengaruh pemberian ekstrak daun Jati Belanda (Guazuma ulmifolia Lamk.) terhadap kualitas sperma mencit (Mus musculus L.) galur Swiss Webster telah dilakukan. Tujuan dari penelitian ini adalah untuk menganalisis pengaruh pemberian ekstrak daun Jati Belanda terhadap kualitas sperma mencit yang terdiri dari konsentrasi sperma (x105/ml suspensi semen), motilitas sperma (%), kecepatan sperma (µm/detik) dan abnormalitas sperma (%). Rancangan Acak Lengkap (RAL) dengan empat kali pengulangan dan enam dosis perlakuan (0,00 g/BB/hari; 0,05 g/BB/hari; 0,10 g/BB/hari; 0,15 g/BB/hari; 0,20 g/BB/hari; 0,25 g/BB/hari) digunakan dalam penelitian ini. Mencit jantan usia empat bulan diberi perlakuan ekstrak daun Jati Belanda dengan cara gavage selama 14 hari. Pada hari ke-15 mencit dimatikan dengan cara dislokasi leher kemudian dipisahkan alat reproduksinya untuk mengamati sperma dalam semen dari kauda epididimis. Hasil menunjukkan pemberian ekstrak daun Jati Belanda signifikan dalam menurunkan konsentrasi sperma. Adapun konsentrasi sperma terendah berada pada dosis pemberian ekstrak daun Jati Belanda sebesar 0,25 g/BB/hari (11,70x105/ml suspensi semen). Persentase rata-rata abnormalitas sperma sekunder meningkat signifikan dengan abnormalitas tertinggi pada pemberian dosis ekstrak daun Jati Belanda 0,25 g/BB/hari (62,38%). Persentase motilitas sperma menunjukan hasil yang tidak signifikan, namun kecepatan gerak sperma menurun secara signifikan. Kecepatan sperma terendah terdapat pada kelompok 0,25 g/BB/hari (65,23 µm/detik). Berdasarkan hasil penelitian tersebut dapat disimpulkan bahwa pemberian ekstrak daun Jati Belanda dapat menurunkan kualitas sperma mencit (konsentrasi sperma, abnormalitas sperma, dan kecepatan gerak sperma). Swiss Webster strains has done.The purpose of this study was to analyze the effect of giving the Bastard Caddar leaf extract on mice sperm quality. Include sperm concentration (x105/ml cement suspension), sperm motility (%), sperm velocity (μm / sec) and sperm abnormalities (%). Complete Random Draft (CRD) with four repetitions and six-dose treatment (0,00 g/bw/day; 0,05 g/bw/day; 0,10 g/bw/day; 0,15 g/bw/day; 0,20 g/bw/day; 0,25 g/bw/day) used in this study. Four months old male mice were subjected to the Bastard Caddar leaf extract by gavage for 14 days. On day 15 mice were switched off by neck dislocation, then the reproductive organs separated to observe sperm in the semen from cauda epididymis. The results show the Bastard Caddar leaf extract significantly in lowering the concentration of sperm. The sperm concentration was lowest at highest dose of the Bastard Caddar leaf extract, 0,25 g/bw/day (11,70 x105/ml cement suspension). The average percentage of secondary abnormalities in sperm abnormalities increased significantly with the highest dose of the Bastard Caddar leaf extract ,0,25 g/bw/day (62,38%). The percentage of sperm motility showed no significant results, but the velocity of sperm decreased significantly. Lowest sperm velocity found in the highest dose of the Bastard Caddar leaf extract, 0,25 g/bw/day (65,23 μm / sec). Based on these results we can conclude that the Bastard Caddar leaf extract can lower the quality of sperm of mice (sperm concentration, sperm abnormalities, and velocity of sperm).
